红丝线草对肝纤维化大鼠肝组织MMP-1及TIMP-1表达的影响

摘要(Abstract):

目的:观察中药红丝线草对肝纤维化大鼠肝组织中基质金属蛋白酶-1(MMP-1)及基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达的影响。方法:将60只SD大鼠随机分为正常组、模型组、秋水仙碱组、红丝线草高、中、低剂量组共6组,每组10只;除正常组外,其余各组大鼠用二甲基亚硝胺(DMN)制作肝纤维化模型;正常组和模型组大鼠用等体积生理盐水喂养,秋水仙碱组、红丝线草高、中、低剂量组分别给予秋水仙碱0.1 mg/kg和高、中、低剂量红丝线草喂养各组大鼠,4周后处死,取大鼠肝脏,采用免疫组织化学方法检测各组大鼠肝组织中MMP-1及TIMP-1蛋白的表达。结果:模型组MMP-1表达明显低于正常组(t=2.57,P<0.05),TIMP-1表达明显高于正常组(t=7.26,P<0.05);与模型组比较,红丝线草高、中剂量组大鼠肝组织中MMP-1蛋白表达明显升高(t=2.77和2.68,P<0.05),TIMP-1蛋白表达明显降低(t=2.76和3.63,P<0.05);红丝线草低剂量组MMP-1及TIMP-1蛋白表达与模型组比较,差异无统计学意义(t=1.16和1.41,P>0.05);红丝线草高、中剂量组肝组织中MMP-1及TIMP-1蛋白表达与秋水仙碱组比较,差异无统计学意义(t=1.29、2.68、0.05、0.16,P>0.05)。结论:红丝线草抗肝纤维化机制可能与MMP-1及TIMP-1表达水平有关。
Objective: To investigate the effect of Peristrophe japonica on the expression of MMP-1and TIMP-1 in hepatic fibrosis rats induced by Dimethylnitrosamine( DMN). Methods: Sixty SD rats were divided randomly into six groups: normal control group,model group,positive control group( colchicin group),and Peristrophe japonica treated groups including high( group H),middle( group M) and low( group L) doses,with 10 rats in each group. Except normal control group,the other groups were induced to construct hepatic fibrosis model with 0. 5% dimethyl nitrosamine( DMN). After model was constructed,positive control group was fed with 0. 1 mg / kg colchicin; group H,group M and group L were fed with high,middle and low doses of Peristrophe japonica respectively,while normal control group and model group were fed with equal volume of saline. The rats in all groups were executed 4 weeks later,and the expression levels of MMP-1,TIMP-1 in liver tissue of rats were measured by immunohistochemistry. Results: In model group,the expression level of MMP-1 protein was obviously lower than that of normal control group( P < 0. 05),and the expression level of TIMP-1 protein was obviously higher than that of normal control group( P < 0. 05). Compared with those of model group,MMP-1 protein expression levels in groups H and M were significantly higher( t = 2. 77 and2. 68,P < 0. 05),and the TIMP-1 protein expression levels were significantly lower( t = 2. 76 and3. 63,P < 0. 05). In group L,the expression levels of the 2 proteins were not obviously different from those of model group( t = 1. 16 and 1. 41,P > 0. 05). The expression levels of the 2 proteins in groups H and M did not differ significantly from that in group colchicin. Conclusion: The anti hepatic fibrosis mechanism of Peristrophe japonica may relate to the expression of MMP-1 and TIMP-1.
