电针对急性饮酒抑郁大鼠海马TLR2/4及炎性细胞因子表达的影响
The Effect of Electroacupuncture on TLR2/4 and Inflammatory Cytokine Expression in Hippocampus of Rats with Acute Ethanol Consumption Depression

摘要(Abstract):

目的:观察电针对急性酒精灌胃后抑郁大鼠海马TLR2/4及炎性细胞因子表达的影响。方法:将48只SD雄性大鼠,随机分成2组,为生理盐水对照组(Control,生理盐水灌胃)和酒精模型组(Ethanol,50%酒精5 g/kg灌胃),通过强迫游泳实验和旷场实验观察两组的行为学改变,若出现抑郁样症状提示造模成功;再随机从Control组和Ethanol组各抽取12只大鼠,为生理盐水加电针组(Control+EA)和酒精模型加电针组(Ethanol+EA),均予电针刺激(1次/d),分别于实验24 h和72 h取大鼠海马组织,用实时荧光定量逆转录聚合酶链反应检测其中Toll样受体2/4(TLR2/4)、白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)mRNA的表达。结果:急性酒精灌胃24 h和72 h后,与Control组比较,Ethanol组大鼠TLR2/4、IL-1β、IL-6、TNF-αmRNA的表达明显增加,差异有统计学意义(P<0.05),Control+EA组无明显变化,差异无统计学意义(P>0.05);与Ethanol组相比,Ethanol+EA组TLR2/4、IL-1β、IL-6、TNF-αmRNA的表达显著减少,差异有统计学意义(P<0.05);Control+EA与Ethanol+EA两组间比较,差异无统计学意义(P>0.05)。结论:电针能下调急性饮酒后24 h、72 h抑郁样行为大鼠海马组织中TLR2/4、IL-1β、IL-6、TNF-αmRNA的表达,这可能参与了电针抗抑郁的机制。
Objective: To investigate the effect of electroacupuncture on TLR2 /4 and pro-inflammatory cytokine expression in hippocampus of rats with acute ethanol intragastric administration. Method:Forty-eight SD male rats were randomly divided into two groups: control group( rats were given normal saline by gavage) and acute ethanol model group( rats were given 50% ethanol 5 g / kg by gavage).Behavioral changes of rats were observed with forced swimming and open field test,and depression-like symptoms suggested that the model was successful. Then 12 rats from each group were randomly selected as control plus electroacupuncture( EA) stimulation group and ethanol plus EA stimulation group,and the rats were accepted EA stimulation once a day. The hippocampus tissues were obtained at 24 h and 72 h after experiment,the expressions of TLR2 /4,IL-1β,IL-6 and TNF-α mRNA were determined with QRT-PCR. Results: Compared with the control group,the expressions of TLR2 /4,IL-1β,IL-6 and TNF-α mRNA in hippocampus of ethanol model rats increased at 24 h and 72 h after acute ethanol intragastric administration( P < 0. 05),yet the differences in the control + EA group were not significant( P > 0. 05). Compared with ethanol group,the expressions of TLR2 /4,IL-1β,IL-6 and TNF-α mRNA in ethanol + EA treatment group significantly decreased( P < 0. 05). However,no statistical difference was found between control + EA group and ethanol + EA group. Conclusion: Electroacupuncture can decrease TLR2 /4,IL-1β,IL-6 and TNF-α mRNA expression in hippocampus rats with acute ethanol intragastric administration. It may be associated with the mechanism of the antidepressive effects of EA.
