LMP7、TAP2基因多态性与云南汉族非小细胞肺癌的相关性Association between Polymorphism of LMP7 and TAP2 Gene and Non-small Cell Lung Cancer in Yunnan Han Nationality

摘要(Abstract):
目的:探讨低相对分子量蛋白酶体7基因(LMP 7)及抗原处理相关转运体2基因(TAP 2)中的单核苷酸多态性(SNP)与云南汉族人群非小细胞肺癌的相关性。方法:选取云南汉族非小细胞肺癌患者467例作为病例组、同期健康正常人群488例作为对照组,采用Taq Man探针基因分型方法对TAP2基因的SNP位点rs241447及LMP7基因的SNP位点rs2071543进行基因分型,分析rs241447和rs2071543等位基因和基因型在病例组和对照组间的分布差异,分析这两个SNP位点在肺癌不同临床分期的中分布差异。结果:rs241447在对照组和病例组中基因型的分布频率比较,差异有统计学意义(P=0.015);在超显性遗传模式下,rs241447基因型C/C-T/T可能是云南汉族人群非小细胞肺癌的风险因素(OR=1.38,95%CI为1.07~1.78,P=0.014),rs2071543的等位基因和基因型在对照组和病例组中的分布频率比较差异无统计学意义(P>0.05)。结论:TAP2基因中的SNP位点rs241447可能与云南汉族人群非小细胞肺癌的易感性相关。
Objective: To investigate the association of single nucleotide polymorphisms( SNP) in low molecular weight proteasome 7( LMP 7) and antigen-associated transporter 2( TAP 2) with nonsmall cell lung cancer in Yunnan Han population. Methods: 467 patients with non-small cell lung cancer( NSCLC) in Yunnan Han nationality were selected as case group and 488 healthy persons as control group. The SNP locus rs241447 of TAP2 gene and SNP rs2071543 of LMP7 gene were genotyped by Taq Man probe genotyping method. The distribution of rs241447 and rs2071543 alleles and genotypes were analyzed between the case group and the control group. The differences between the two SNP loci in different clinical stages of lung cancer were analyzed. Results: The genotype distribution of rs241447 between control and case groups was compared,and the difference was statistically significant( P = 0. 015). The genotype C/C-T/Tof rs241447 was a risk factor for non-small cell lung cancer in Yunnan Han population( P = 0. 014,OR = 1. 38,95% CI was 1. 07 ~ 1. 78),and there was no significant difference in the distribution frequency of rs2071543 alleles and genotypes between the control group and the case group( P > 0. 05). Conclusion: The SNP locus in TAP2 gene may be associated with the susceptibility of non-small cell lung cancer in Yunnan Han population.
