摘要(Abstract):
目的:探讨苗药防感香囊对小鼠肺组织白介素1受体相关激酶M(IRAK-M)、细胞因子信号抑制因子(SOCS1)基因和蛋白表达的影响。方法:48只小鼠随机分为空白对照组(A组)、单纯香囊持续吸入组(B组)、单纯冷刺激组(C组)、香囊持续吸入加冷刺激组(D组),每组12只,分别给予相应干预,37 d后收集肺组织,采用定量PCR和免疫印迹法检测小鼠肺组织IRAK-M、SOCS1 mRNA和蛋白表达。结果:B、D组小鼠肺组织中IRAK-M mRNA表达水平低下,与A、C组比较差异有统计学意义(P<0.05);B、D组肺组织中SOCS1 mRNA表达水平低于C组(P<0.05);B组SOCS1蛋白表达水平低下,与A、C组比较差异有统计学意义(P<0.05);其余各组IRAK-M的蛋白表达水平,差异无统计学意义(P>0.05)。结论:苗药防感香囊能够下调TLR通路的负性调控因子IRAK-M、SOCS1 mRNA及SOCS1蛋白的表达水平,增强TLR通路介导的天然免疫功能。
Objective: To investigate the effect of Miaoyao anti-influenza sachet( referred to as: Miao Sachet) on lab rat lung IRAK-M,SOCS1 gene and protein expression. Methods: All the rats were divided into 4 groups randomly in equal numbers,control group( Group A); inhaling Miao Sachet group( Group B); cold stimulating group( Group C); inhaling Miao Sachet and cold stimulating group( Group D). Lung tissues were collected 37 days later,quantitative PCR and western blot were adopted to test IRAK-M,SOCS1 gene and protein expression. Results: The expression of IRAK-M and SOCS1 gene of B and D groups was lower than that of A and C groups,the differences were statistically significant( P < 0. 05); the expression of SOCS1 gene of B and D groups was lower than C group( P <0. 05),the expression of SOCS1 gene of B group was lower than A and C groups,differences were statistically significant( P < 0. 05); comparing expressions of IRAK-M protein with each other,there was no statistical significance( P > 0. 05). Conclusions: Expression of IRAK-M、SOCS1 expression could be regulated by Miao Sachet with continued inhaling.
