高效液相色谱法测定清热丸中黄芩苷的含量

摘要(Abstract):

目的:建立高效液相色谱法(HPLC)测定清热丸中黄芩苷的含量。方法:采用反相高效液相色谱法,C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-0.4%磷酸溶液(51∶49),检测波长为277 nm,流速为1.0 ml/min,柱温为40℃。结果:黄芩苷在0.086 5~0.770 4μg与峰面积呈良好的线性关系(r=0.999 9),该制剂中黄芩苷平均回收率为99.47%,RSD为1.08%(n=6)。结论:所建立的方法简便易行、结果可靠、重复性好,可用于清热丸中黄芩苷的含量测定。
Objective: To establish a method for determination of baicalin in Qingre pills with high performance liquid chromatography(HPLC).Methods: The HPLC method was carried on C18(4.6 mm×250 mm,5 μm)column using methanol-0.4% phosphoric acid(51∶49,V∶V) as mobile phase,and detection wavelength was 277 nm.The flow rate was 1.0 ml/min and the column temperature was 40 ℃.Results: Baicalin amount presented a fine linear relationship with peak area in the range of 0.086 5~0.770 4 μg(r=0.999 9).The average recovery was 99.47%(n=6),and PSD was 1.08%(n=6).Conclusion: The method is convenient,precise and reliable for determining the content of baicalin in Qingre pills.
