上皮性卵巢癌血管内皮生长因子表达及血管生成的研究

摘要(Abstract):

目的:比较上皮性卵巢癌组织与癌周正常卵巢组织中的微血管密度(microvessel density,MVD)及血管内皮生长因子(vascular endothelial growth factor,VECF)的表达,探讨血管生成与上皮性卵巢癌的生长、临床分期、细胞学分级、转移、治疗、预后的关系。方法:用免疫组织化学SP法(过氧化物酶标记链霉菌抗生物素蛋白法),对上皮性卵巢癌的组织切片行Ⅷ因子相关抗体和VEGF抗体免疫组织化学染色。结果:上皮性卵巢癌组织MVD及VEGF表达明显高于癌周正常卵巢组织,P<0.001;有淋巴结转移患者的MVD高于无淋巴结转移患者,P<0.05;癌组织中的VEGF表达百分率与MVD之间呈直线相关,相关系数0.3811,P<0.05;癌周正常卵巢组织内的VEGF表达与MVD没有相关性,相关系数为-0.1902,P>0.1。多元分析提示MVD与临床分期、细胞学分级有关。结论:MVD和VEGF表达可作为反映上皮性卵巢癌血管生成的指标。血管生成与上皮性卵巢癌的生长、转移、预后有密切关系。寻找抑制肿瘤血管生成的药物可为抗肿瘤治疗开辟新途径。
Objective: To investigate the correlation between angiogenesis and FIGO stage, cytologic grade, metastasis, prognosis and treatment of epithelial ovarian carcinoma. The differnces of microves-sel density ( MVD) and expression of vascular endothelial growth factor (VEGF) in carcinoma tissues and neighbouring normal ovarian tissues of primary epithelial ovarian carcinoma were studied. Methods : 32 samples of tissue from patients with primary epithelial ovarian carcinoma were collected during ovarian malignant tumor debulking at the affiliated hospital of Guiyang Medical College during 1979-1994. Sections were immunohistochemically stained with commercially available antibody to VEGF and factor Ⅷ related antigen by streptoaidin-biodin peroxidase technique ( SP) , respectively. MVD and expression of VEGF were assessed with light microscope over carcinoma tissues and neighbouring normal ovarian tissues. Results: High MVD and high expression of VEGF were mainly found within carcinoma tissues of primary epithelial ovarin carcinoma compared with neighbouring normal ovarian tissues (P <0. 001 ) . Within carcinoma tissues, patients with melastases of omental lymph-nodes had higher MVD and expression of VEGF than patients without metastases. Linear regression analysis suggested expression of VEGF correlated to MVD within carcinoma tissues, ( r=0. 381 1 ,P <0. 05 ) . No correlation were found between expression of VEGF and MVD within neighbouring normal ovarian Lissues, ( r = -0. 1902, P >0. 1). Multiple linear correlative analysis showed that MVD correlated to FIGO stage and cytological grade within carcinoma tissues . Conlusions: This study shows angiogenesis detected indicated by VEGF correlates closely to growth and metastasis of primary epithelial ovarian carcinoma. This correlation suggests MVD might be an indicator for prognosis. Microvessels within epithelial ovarian carcinoma might be a promising therapeutic target.
