卡维地洛对ox-LDL诱导的人主动脉内皮细胞氧化损伤的保护作用Protective Effect of Carvedilol against Ox-LDL Induced Oxidative Injury of Human Aortic Endothelial Cells in vitro

摘要(Abstract):
目的:研究卡维地洛(CAR)对氧化低密度脂蛋白(ox-LDL)诱导的人主动脉内皮细胞(HAECs)损伤的保护作用。方法:体外培养HAECs,分为对照组、模型组、CAR高剂量组(CAR H组)和CAR低剂量(CAR L组),即对照组(无血清DMEM)、模型组(200 mg/L ox-LDL 100μL)、CAR高剂量组(CAR H组,200 mg/L oxLDL及1×10-5mol/L CAR各100μL)及CAR低剂量组(CAR L组,200 mg/L ox-LDL及1×10-6mol/L CAR各100μL),后两组CAR预保护1 h后,加入ox-LDL作用24 h;采用MTT法分析细胞存活率,吉姆萨(Giemsa)染色后观察HAECs形态;酶联免疫吸附法(ELISA)检测培养液中一氧化氮(NO)含量及乳酸脱氢酶(LDH)外漏量。结果:模型组与对照组相比,细胞发生严重的病理形态学改变,细胞存活率及NO含量显著降低,LDH外漏量显著升高(P<0.05),CAR H组和CAR L组细胞接近对照组,细胞存活率及NO含量较模型组显著提高,LDH外漏量显著降低(P<0.05)。结论:CAR能减少ox-LDL诱导的HAECs损伤,其作用可能与NO系统有关。
Objective: To investigate the protective effect of carvedilol(CAR) against oxidized low density lipoprotein( ox-LDL) induced human aortic endothelial cells( HAECs) injury. Methods:HAECs were cultured in vitro,randomly divided into four groups: control group( serum free DMEM),model group(200 mg / L ox-LDL 100 μL),carvedilol high dose group( CAR H group,200 mg / L oxLDL and 1 × 10-5mol / L CAR) and carvedilol low dose group( CAR L group,200 mg / L ox-LDL and1 × 10-6mol / L CAR). Two latter groups preincubated with carvedilol for 1 h and add ox-LDL for24 h; adopting MTT assay to analyze cell viability,HAECs morphology was observed by Giemsa staining. The contents of nitric oxide( NO) and the leakage of lactate dehydrogenase( LDH) were detected by enzyme-linked immunosorbent assay( ELISA). Results: Comparing model group with control group,cells underwent severe pathomorpholohy change,cell viability and NO content decreased significantly,LDH leakage increased significantly( P < 0. 05),cells of CAR H group and CAR L group were similar to control group,cell viability and NO content increased more than model group,LDH leakage decreased significantly( P < 0. 05). Conclusion: Carvedilol could ameliorate the HAECs injury induced by ox-LDL,which maybe involve in NO system.
