蜘蛛香总缬草三酯的平衡溶解度、表观油水分配系数和稳定性测定

摘要(Abstract):

目的:测定蜘蛛香总缬草三酯的平衡溶解度及表观油水分配系数,考察其在水及酸、碱缓冲液中的稳定性。方法:采用紫外分光光度法测定蜘蛛香总缬草三酯在10种有机溶剂(甲醇、乙腈、乙醇、丙酮、异丙醇、乙酸乙酯、正辛醇、二氯甲烷、石油醚及正己烷)的平衡溶解度,应用摇瓶法测定蜘蛛香总缬草三酯在正辛醇饱和的水及缓冲溶液中的表观油水分配系数,利用紫外分光光度法考察蜘蛛香总缬草三酯在水及酸碱缓冲液中的稳定性。结果:蜘蛛香总缬草三酯在异丙醇、丙酮、乙醇等有机溶剂中溶解度大,在正辛醇饱和的水中的表观油水分配系数为494.667(lg P=2.694),在水及酸、碱缓冲液中均不稳定。结论:蜘蛛香总缬草三酯水溶性及稳定性差,脂溶性好。
Objective: To determine equilibrium solubility and apparent oil/water partition coefficient of total valepotriates from Valeriana jatamansi,and to investigate its aqueous and p H chemical stability. Methods: Ultraviolet spectrophotometry was employed to determine the equilibrium solubility of total valepotriates from Valeriana jatamansi in 10 kinds of organic solvents,while the apparent oil/water partition coefficient for the n-octanol/water and buffer salt solution system was determined by shaking flask method. The aqueous and p H chemical stability of total valepotriates was determined by ultraviolet spectrophotometer method. Results: The equilibrium solubility of total valepotriates was high in organic solvents such as isopropyl alcohol,acetone,and ethanol,and its partition coefficient was494. 667( lg P = 2. 694) in the n-octanol/water system. It was unstable in water and buffer solution.Conclusion: Total valepotriates from Valeriana jatamansi has poor solubility in water and poor stability. While it has high lipophilicity.
