摘要(Abstract):
目的:探讨三结构域蛋白31(TRIM31)对胰腺癌侵袭转移的影响及其机制。方法:利用基因表达数据库(GEO)获得TRIM31在胰腺癌组织、癌旁组织以及正常胰腺组织中的表达,选取两株胰腺癌细胞Panc-1及MIAPa Ca-2分别分为TRIM31沉默组(实验组)和空载体组(阴性对照组),通过Transwell小室细胞迁移和划痕实验观察TRIM31对胰腺癌细胞侵袭和迁移能力的影响,Western blot实验检测上皮-间充质细胞转化(EMT)相关钙黏附蛋白E(E-cadherin)和波形蛋白(Vimentin)的表达水平。结果:正常胰腺组织中TRIM31 mRNA相对表达量明显低于胰腺癌组织,癌旁组织中TRIM31 mRNA相对表达量明显低于癌组织,差异有统计学意义(P<0.05);小室细胞迁移和侵袭实验显示,实验组Panc-1及MIA Pa Ca-2迁移和侵袭细胞数明显低于阴性对照组,差异有统计学意义(P<0.05);沉默TRIM31表达后,Panc-1及MIAPa Ca-2细胞中E-cadherin蛋白表达量明显升高,Vimentin蛋白表达量明显降低。结论:沉默TRIM31的表达可能通过抑制胰腺癌细胞的EMT过程,进而抑制其侵袭迁移。
Objective: To investigate the effect of tripartite motif-containing protein 31( TRIM31) on the invasion and migration of pancreatic ductal adenocarcinoma( PDAC) cells and its mechanism.Methods: The expression relationship of TRIM31 in pancreatic ductal adenocarcinoma,adjacent normal tissue and normal tissue samples were determined by data analysis of GEO. Two strains of Panc-1 and MIAPa Ca-2 were selected as TRIM31 silencing group( experiment group) and Lip-null group( negative control group). Transwell assay and scratch assay were conducted to observe the ability of TRIM31 on pancreatic ductal adenocarcinoma cell invasion and migration; Western blot was conducted to determine expression of EMT E-cadherin and Vimentin. Results: The TRIM31 mRNA relative expression in the pancreatic ductal adenocarcinoma were significantly higher than those in the normal tissue and adjacent normal tissue,differences were statistically significantly( P < 0. 05). Invasion experiment showed that invasion and migration cell numbers of Panc-1 and MIA Pa Ca-2 in experiment group were significantly lower than negative control group,differences were statistically significantly( P <0. 05). After TRIM31 silencing,the expression level of E-cadherin increased and Vimentin downregulated in Panc-1 and MIAPa Ca-2 cells. Conclusion: TRIM31 silencing inhibits the invasion and migration of pancreatic cancer via regulating the process of epithelial-mesenchymal transition.
