细胞培养密度对乳腺癌4T1细胞增殖、迁移、黏附及浸润能力的影响

摘要(Abstract):

目的:探讨细胞培养密度对乳腺癌4T1细胞增殖、迁移、黏附及浸润能力的影响。方法:取乳腺癌4T1细胞,培养至汇合度为60%(低密度培养组,sparse组)和90%(高密度培养组,dense组),两组取相同数量的4T1细胞,采用克隆实验检测两组细胞的克隆形成数目,采用划痕实验和Transwell小室跨膜迁移实验检测4T1细胞的迁移率,采用黏附实验检测细胞的黏附率,采用Transwell-Matrigel细胞浸润实验检测4T1细胞侵袭能力。结果:克隆实验结果显示,与sparse组比较,dense组的4T1细胞形成克隆数目明显增多(P <0.01),且形成的单个克隆面积更大;划痕实验结果显示,与sparse组比较,dense组4T1细胞划痕愈合率显著升高(P <0.01); Transwell小室跨膜迁移实验结果显示,dense组4T1细胞迁移率明显高于sparse组(P <0.01);黏附实验结果显示,dense组4T1细胞的黏附率明显高于sparse组(P <0.05); Transwell-Matrigel实验结果显示,dense组4T1细胞的浸润能力明显高于sparse组(P <0.01)。结论:高密度培养能够明显增强乳腺癌4T1细胞的增殖、迁移、黏附和浸润能力。
Objective: To investigate the effect of cell culture density on the proliferation,migration,adhesion and invasion of breast cancer 4 T1 cell line. Methods: The same numbers of breast cancer4 T1 cells derived from the confluence of 60%( low-density group,sparse gruop) and 90%( highdensity group,dense group) were seeded for clonogenic assay to detect cell proliferation,for woundhealing assay and Transwell migration assay to measure cell migration rates,for adhesion test to determine cell adhesion rate,and for Transwell-Matrigel invasion assay to detect cell invasion ability.Results: Compared with sparse group,the number and the size of the colonies,the wound-healing rate,the migration rate significantly increased in dense group( P < 0. 01). Similarly,the adhesion test showed that the adhesion rate was higher in dense group than that in the sparse group( P < 0. 05).Moreover,Transwell-Matrigel assay showed the invasive ability 4 T1 cells from high density group was higher than that of low-density group( P < 0. 01). Conclusion: High-density culture can significantly enhance the proliferation,migration,adhesion and invasion potential of breast cancer 4 T1 cells.
