摘要(Abstract):
目的:探讨Otos基因的表达对碱性成纤维细胞生长因子(bFGF)的调控作用,以揭示Otos基因和bFGF在耳聋病理生理过程中的联系和作用。方法:用含有大鼠Otos基因的腺病毒载体Ad-Otos转染培养的大鼠耳蜗螺旋韧带成纤维细胞,应用RT-PCR技术从转染细胞中检测Otos基因和bFGF基因的表达,用Westernblot方法检测转染细胞中的bFGF蛋白的表达。结果:经过转染Ad-Otos后,成纤维细胞中的bFGF显著上调。结论:Otos基因的表达可能对bFGF表达有影响,并有可能对内耳有保护作用。
Objective:To investigate if there might be any influence of Otos gene expression on the regulation of basic fibroblast growth factor(bFGF),and to reveal the possible relationship and interactions between them and the path-physiological course of epicophosiss.Methods:Cultured spiral ligament fibrocytes of SD rats were transfected with different expression vectors including Ad-Otos which contains Otos gene,AD-GFP containing green flourescence protein gene,and DMEM respectively,and,then,mRNA of Otos and bFGF as well as protein of bFGF were assessed with RT-PCR and Western blot.Results:Compared with those in the control cells,mRNA expression of Otos and mRNA and expressed protein of bFGF in the fibrocytes transfected with Ad-Otos evidently increased.Conclusions:Elevated expression of Otos gene might have an influence on expression of bFGF and have potential protective effects to inner ear.
