骨康胶囊组分酢浆草和芭蕉根对家兔骨折愈合的作用及机制Effects of oxalis corniculata and musa basjoo sieb of gukang capsule on fracture healing in rabbits and its mechanism

摘要(Abstract):
目的 探讨骨康胶囊组分酢浆草、芭蕉根对家兔骨折愈合的作用及机制。方法 35只家兔随机均分为正常组(仅蒸馏水灌胃)、模型组(蒸馏水10 mL/kg)、接骨七厘片组(0.140 g生药/kg)、酢浆草组(0.224 g生药/kg)及芭蕉根组(0.336 g生药/kg),后4组家兔构建左侧桡骨骨折模型后、按体质量10 mL/kg灌胃相应药物,对照组灌胃蒸馏水,连续30 d;末次灌胃结束后,取各组家兔心脏血检测碱性磷酸酶(ALP)、钙(Ca)、磷(P)的含量,检测血液流变学参数(全血低切黏度、全血中切黏度、全血高切黏度、血浆黏度、红细胞压积及血沉);处死各组家兔,取左侧桡骨骨折部位切片,苏木精-伊红染色(HE)法观察组织学特征。结果 与模型组家兔比较,酢浆草、芭蕉根组家兔ALP水平明显增高(P<0.01),酢浆草组和芭蕉根组家兔全血中切、高切黏度降低(P<0.05或P<0.01);与模型组比较,酢浆草组和芭蕉根组家兔骨折部位可见大量纤维组织、成骨细胞,断端和骨膜下出现较多类骨质。结论 骨康胶囊组分酢浆草和芭蕉根可促进家兔骨折愈合,其机制可能与上调ALP、降低血液黏度及改善血液流变性有关。
Objective To investigate the effects and mechanism of oxalis corniculata and musa basjoo sieb of gukang capsule on fracture healing in rabbits.Methods Thirty-five rabbits were randomly divided into normal group(intragastric administration of distilled water only), model group(distilled water intragastric administration 10 mL/kg), jieguqili tablet group(0. 140 g crude drug/kg), oxalis corniculata(0. 224 g crude drug/kg), and musa basjoo sieb groups(0. 336 g crude drug/kg). After the fracture model of left radius was established in the latter 4 groups except, intragastric administration with distilled water at 10 mL/kg of weight in suspension was given for consecutive 30days, control group was given distilled water gastric administration for the same time. After the last administration,the contents of alkaline phosphatase( ALP), calcium( Ca), and phosphorus( P) in serum of rabbits in each group were determined. Detecting rheology parameters in observation groups(including whole blood low shear viscosity, whole blood midium shear viscosity, whole blood high shear viscosity, plasma viscosity, hematocrit, and erythrocyte sedimentation rate) of rabbits in each group. After death, the left radial bone fracture sites of rabbits in each group were selected and sectioning was conducted, hematoxylin-eosin staining(HE) was used to observe the histopathological characteristics.Results The results of serum ALP level in oxalis corniculata group and musa basjoo sieb group were obviously increased than that in model group(P< 0. 01). In oxalis corniculata group and musa basjoo sieb group, the viscosity of medium cut and high cut in whole blood was decreased(P< 0. 05 orP< 0. 01). Pathological results showed that compared with the model group, there were more fibrous tissues and osteoblasts at the fracture site in oxalis corniculata group and musa basjoo sieb group; and more osteoid at the fracture end and subperiosteum in these two groups.Conclusion The components of gukang capsule, oxalis corniculata, and musa basjoo sieb can promote fracture healing in rabbits, and the mechanism may be related to the up-regulation of ALP, the decrease of blood viscosity, and the improvement of blood rheology.
