几种消毒剂对蚋卵原代细胞培养效果的比较Comparison of Sterilizing Effects of Several Disinfectants on Blackfly Eggs in Primary Cell Culture

摘要(Abstract):
目的:探索适宜于蚋卵(胚胎)原代细胞培养的消毒方法。方法:70%乙醇、2.8%NaClO、15%甲酚皂、0.1%HgCl2及1%过氧乙酸5种消毒剂按不同消毒时间相互搭配,结合使用抗生素PBS液对蚋卵进行消毒处理。结果:70%乙醇10 min、1%过氧乙酸15 min、2.8%NaClO5 min 3种消毒剂的相互搭配,结合使用了抗生素PBS液,使得原代细胞培养污染得到有效控制,细胞生长和增殖状态较好。结论:70%乙醇10 min、1%过氧乙酸15 min、2.8%NaClO 5 min联合使用是蚋卵原代细胞培养的一种安全有效的消毒方法。
Objective: To explore disinfecting methods for primary cell culture of blackfly eggs(embryos).Methods: Different combination of 70% ethanol,2.8% NaClO,15% cresol soap,0.1%HgCl2 or 1% peroxyacetic acid respectively with antibiotics PBS,and combination of their different montages were tried to disinfect eggs(embryos) for different minutes,and the results were compared.Results: Combination of 70% ethanol treating for 10 min,1% peroxyacetic acid for 15 min,and 2.8% NaClO for 5 min plus treatment of antibiotics PBS controlled contamination effectively and cells of the first generation grew well in vitro.Conclusions: The joint use of 70% ethanol for 10 min,1% peroxyacetic acid for 15 min,and 2.8% NaClO for 5 min is a safe and effective scheme for blackfly eggs(embryos) disinfection in primary cell culture.
