CENP-A在口腔鳞状细胞癌中的表达及意义Expression and Clinical Significance of Centromere Protein A in Oral Squamous Carcinoma Tissues

摘要(Abstract):
目的:探讨着丝粒蛋白A(CENP-A)在口腔鳞状细胞癌(OSCC)组织中的表达及其与OSCC患者临床病理因素之间的关系。方法:收集32例OSCC组织及同期20例口腔正常组织,采用免疫组织化学染色SP法及实时荧光定量PCR(q PCR)检测CENP-A蛋白及mRNA的表达,观察不同临床病理特征的OSCC组织中CENPA基因表达的差异。结果:与口腔正常组织比较,CENP-A蛋白和mRNA在OSCC组织中的表达均明显升高,差异有统计学意义(P<0.05);OSCC组织中的CENP-A的蛋白和mRNA表达随着肿瘤体积增大、淋巴转移而升高(P<0.05);但与患者年龄、性别及肿瘤分化无关(P>0.05)。结论:OSCC组织中CENP-A蛋白和mRNA的表达明显高于正常组织,其表达与肿瘤体积大小、淋巴转移有关。
Objective: To explore the expression level of centromere protein A(CENP-A) in oral squamous carcinoma tissues(OSCC) and its relationship with various clinical pathological factors of patients with OSCC. Methods: 32 cases of OSCC tissues and 20 cases of normal oral tissues at the same period were collected. The immunohistochemistry SP method and real-time quantitative PCR was adopted to detect the protein and mRNA expressions of CENP-A. The difference of CENP-A gene expression in OSCC tissues with different clinicopathological features was observed. Results: Compared with normal oral tissues,the expression of CENP-A protein and mRNA in OSCC tissue was significantly increased,and the difference was statistically significant(P < 0. 05). The expression of CENP-A protein and mRNA in OSCC increased with the increase of tumor volume and lymph node metastasis(P <0. 05) but was not related to age,sex and tumor differentiation and stage(P > 0. 05). Conclusion:The protein and mRNA expression of CENP-A in OSCC tissues is significantly higher than that in normal tissues,and the expression is related to tumor size and lymph node metastasis.
