芍药苷对人胰腺癌Panc-1细胞系增殖和凋亡的影响

摘要(Abstract):

目的:探究芍药苷(PF)对人胰腺癌Panc-1细胞增殖及凋亡的影响。方法:将人胰腺癌Panc-1细胞分为对照组(0μmol/L PF)和12.5、50、200及800μmol/L PF组,用细胞增殖实验(CCK8)和平板克隆实验检测PF对人胰腺癌Panc-1细胞增殖的影响,用Annexin V-FITC/PI双染试剂染色实验检测人胰腺癌Panc-1细胞凋亡,免疫印迹实验检测PF对人胰腺癌Panc-1细胞Caspase-3及Bcl-2蛋白表达水平的影响。结果:CCK8实验显示Panc-1细胞增殖能力随药物浓度的增加而减少(P<0.05),平板克隆实验显示细胞集落数量随药物浓度的增加而减少(P<0.05),Annexin V-FITC/PI双染试剂染色实验显示细胞凋亡发生率随药物浓度的增加而增加(P<0.05);Western blot实验结果显示Caspase-3、Cleave caspase3的表达量随PF浓度的增加而增加(P<0.05),Bcl-2与其相反。结论:PF可以抑制人胰腺癌Panc-1细胞的增殖、促进其凋亡,其机制可能是激活了Caspase途径。
Objective: To investigate the effect of Paeoniflorin( PF) on the proliferation and apoptosis of pancreatic cancer Panc-1 cells. Methods: Pancreatic cancer Panc-1 cells were divided into 0,12. 5,50,200 and 800 μmol/L PF group; The effect of different concentrations of Paeoniflorin on the proliferation of Panc-1 by CCK-8 assay and Plate cloning. Apoptosis of Panc-1 was detected by Annexin V-FITC/PI double staining test and Western blot was adopted to detect effect of PF on Panc-1 Caspase-3 and Bcl-2 protein expression. Results: CCK8 experiments indicated that proliferation of Panc-1 reduced with the increase of concentration level of PF( P < 0. 05); plate cloning experiment showed that colony formation number reduced with the increase of concentration level of PF( P <0. 05); Annexin V-FITC/PI double staining test indicated that apoptosis occurrence rate increased with the increase of concentration level of PF( P < 0. 05); Western blot results showed that Caspase-3 and Cleave caspase3 expression increased with the increase of PF concentration( P < 0. 05),but Bcl-2 contrast with it. Conclusion: Paeoniflorin can inhibit the cell proliferation and promote cell apoptosis on human pancreatic cancer cell line Panc-1 cells,its mechanisms is to activate key enzyme Caspase-3 pathway.
