人表皮黑素细胞与角质形成细胞体外共培养模型的构建

摘要(Abstract):

目的:体外构建人表皮黑素细胞与角质形成细胞直接接触的共培养模型。方法:胰酶消化包皮的表皮,分别获取纯化的角质形成细胞和黑素细胞,将第3代角质形成细胞接种于培养瓶中,48 h后以1∶10(表皮黑素细胞∶角质形成细胞)的比例接种黑素细胞。结果:黑素细胞与角质形成细胞混合培养5 d后,培养瓶瓶底培养物逐渐变为浅灰色,继续培养,颜色逐渐加深;倒置显微镜下观察到黑素细胞与角质形成细胞共同存活,细胞生长良好,并通过树突发生联系。结论:成功构建了黑素细胞与角质形成细胞直接接触的混合培养模型,为进一步研究色素问题奠定了基础。
Objective:To construct a co-cultivating model of keratinocytes(KCs) and melanocytes(MCs) in vitro.Methods: Single-cell suspensions were obtained by treating the isolated preputial epidermis with pancreatin,then KCs and MCs were respectively purified at the second passage.The third passage of KCs was transplanted into culture flask and incubated for 48 hours.MCs were then added into the culture flask making a ratio of 1∶10 of MCs:KCs.Results: The model showed a light gray color in 5 days of cultivating,and the color became darker gradually when cultivating continued.Alive MCs and KCs were observed in the co-culture system and dendritic contact between them was found with inverted microscopy.Conclusion: The co-cultivating model of MCs and KCs has been constructed successfully,which provides basis for further study.
