妇科再造丸含药血清对子宫肌瘤细胞增殖和凋亡的影响
Effect of Fu-Ke-Zai-Zao Pill Medicated Serum on Proliferation and Apoptosis of Human Uterine Leiomyoma Cells

摘要(Abstract):

目的:观察妇科再造丸(FKW)含药血清对体外培养子宫肌瘤细胞增殖和凋亡的影响及对人第10号染色体缺失的磷酸酶及张力蛋白同源基因(PTEN)蛋白表达的调控作用。方法:取手术切除的子宫肌瘤标本,分离人子宫肌瘤原代细胞培养至3~4代;雌性SD大鼠灌喂高、中、低剂量FKW药液3 d,取大鼠血清作用于培养的3~4代的子宫肌瘤细胞,采用MTT法检测细胞的增殖活力,流式细胞术检测细胞的凋亡率,免疫细胞化学法测定PTEN蛋白表达情况。结果:成功进行人子宫肌瘤细胞原代培养,培养至3~4代的子宫肌瘤细胞纯度接近100%;FKW各剂量组含药血清作用72 h后能显著降低子宫肌瘤细胞的增殖活力(P<0.05),提高其凋亡率(P<0.01),并且能上调子宫肌瘤细胞PTEN的表达(P<0.01)。结论:FKW含药血清抑制子宫肌瘤细胞增殖,诱导其凋亡的作用可能是通过上调PTEN表达实现的。
Objective: To observe the regulating effect of medicated serum of Fu-Ke-Zai-Zao pill(FKW) on proliferation and apoptosis of human uterine leiomyoma cells cultured in vitro and the regulation of PTEN protein expression.Methods:Hysteromyoma specimens were collected,the uterine leiomyoma cells were separated and cultured to 3-4 generations.Female SD rats were fed with high,medium and low dose of FKW drug solution for 3 days,and serum of rats was taken and applied to cultured uterine leiomyoma cells for 3 to 4 generations.MTT method was adopted to detect cell proliferation activity,flow cytometry was used to detect cell apoptosis rate,and PTEN protein expression was determined by immunocytochemistry.Results: The primary culture of human uterine leiomyoma cells was successfully carried out,and the purity of 3 to 4 generations uterine leiomyoma cells were close to 100%.After 72 h treatment with serum of each dose group of FKW,the proliferation activity of uterine fibroid cells was significantly reduced(P<0.05),the apoptosis rate was increased(P<0.01),and the expression of PTEN of uterine leiomyoma cells was upregulated(P<0.01).Conclusion The effect of FKW medicated serum on inhibiting the proliferation of uterine leiomyoma cells and inducing their apoptosis may be realized by up-regulating the expression of PTEN.
