FZD3基因和蛋白在大肠癌中的表达Expression of FZD3 Gene and Protein in Colorectal Cancer

摘要(Abstract):
目的:探讨WNT信号通路受体FZD3mRNA及蛋白在大肠癌细胞及组织中的表达。方法:取正常大肠细胞株CCD18-Co、大肠癌细胞株SW480和SW620,以WNT信号通路PCR芯片筛选大肠癌细胞株中差异性表达的基因,并选择FZD3基因采用Taqman PCR法进行验证其在大肠癌细胞中的高表达,同时比较FZD3蛋白在大肠癌组织、腺瘤组织及癌旁正常组织的表达。结果:与大肠正常细胞株CCD18-Co比较,19个基因在大肠癌细胞株SW480和SW620中表达上调,其中FZD3基因为CCD18-Co的195倍和105倍;验证结果显示,与大肠正常细胞株CCD18-Co FZD3比较,FZD3 mRNA在SW480和SW620细胞株中较中升高820倍和622倍,验证了FZD3mRNA在大肠癌细胞株中的高表达;免疫组化结果显示,FZD3蛋白在大肠癌组织中表达为100%,大肠腺瘤中的表达为88%,而癌旁正常组织中仅为17.5%,与癌旁正常组织相比,FZD3蛋白在大肠癌组织中免疫组化分数高6.6倍(P<0.01);FZD3蛋白的高表达与肠癌TNM分期显著相关(P<0.005)。结论:FZD3可能在大肠癌的发生、发展中起到潜在癌基因的作用。
Objective: To explore mRNA and protein expression of FZD3 receptor for WNT signaling pathway in colorectal cancer( CRC) cell lines and tissue and its clinical significance. Methods: Normal colorectal cell line CCD18-Co,colorectal cancer cell line SW480 and colorectal cancer cell line SW620 were selected in this study. The differentially expressed genes in colorectal cancer cell lines and normal colorectal cell line were screened by WNT signaling pathway PCR array,and FZD3 gene was chosen to validate the overexpression in CRC cell lines by using Taqman PCR assay. Meanwhile,FZD3 protein expression profile was also detected and compared between colorectal cancer tissue,adenoma tissue and adjacent normal tissue. Results: Compared with normal colorectal cell line CCD18-Co,nineteen genes up-regulated their expression in CRC cell lines SW480 and SW620,of which FZD3 gene expression was up-regulated by 195 and 105 folds in SW480 and SW620,respectively. Identification results showed compared with normal colorectal cell line CCD18-Co,FZD3 mRNA up-regulated its expression in SW480 and in SW620 by 820 folds and 622 folds,respectively. Immunohistochemical results showed that the expression of FZD3 protein in colorectal cancer tissue was 100%,the expression in colorectal adenoma was 88%,and the expression in the adjacent normal tissues was only17. 5%. There was a 6. 6-fold up-regulation of FZD3 protein expression in CRC tissue compared with in adjacent normal tissue( P < 0. 01). The high expression of FZD3 protein was significantly correlated with the TNM staging of colorectal cancer( P < 0. 005). Conclusion: FZD3 may play a role in CRC tumorigenesis and development as a potential oncogene.
