人参皂苷Rg1对心肌纤维化模型小鼠TLR4/MyD88通路的影响
Effects of ginsenoside Rg1 on myocardial fibrosis in mice via TLR4/MyD88 pathway

摘要(Abstract):

目的 观察人参皂苷Rg1(GRg1)对心肌纤维化模型小鼠Toll样受体4/髓样分化因子88(TLR4/MyD88)信号通路的影响。方法 30只雄性C57BL/6J小鼠随机分为对照组、模型组和人参皂苷Rg1干预组,造模成功后取各组小鼠心肌组织,苏木精-伊红染色法(HE)、马松染色(Masson)染色观察心肌组织学改变,免疫印迹试验(WB)检测各组小鼠心肌组织中TLR4/MyD88信号通路TLR4、MyD88、Bax及Bcl-2等凋亡蛋白的表达,聚合酶链式反应(PCR)检测心肌组织中TLR4、MyD88、Bax、Bcl-2等基因表达。结果 HE染色和Masson染色结果均显示,对照组小鼠心肌细胞形态规则正常,细胞间排列顺序整齐;ISO组中纤维化后的小鼠心肌细胞形态和心肌细胞不规则;WB结果表明,与对照组比较,ISO组小鼠心肌组织中TLR4、MyD88、Bax蛋白表达明显升高,Bcl-2表达降低,差异有统计学意义(P<0.05);与ISO组比较,Rg1组小鼠心肌组织中TLR4、MyD88、Bax的表达明显降低,Bcl-2表达升高,差异有统计学意义(P<0.05);PCR结果表明,与对照组比较,ISO组小鼠心肌组织中TLR4、MyD88、Bax的mRNA明显升高,Bcl-2mRNA表达降低,差异有统计学意义(P<0.05);与ISO组比较,Rg1组小鼠心肌组织中TLR4、MyD88、Bax的mRNA明显降低,Bcl-2mRNA表达升高,差异有统计学意义(P<0.05)。结论 人参皂苷Rg1可能通过影响心肌纤维化模型小鼠心肌细胞TLR4/MyD88信号通路活化,延缓心肌纤维化进程,发挥心脏保护作用。
Objective To observe the effects of ginsenoside Rg1 on TLR4/MyD88 signaling pathway in model mice with myocardial fibrosis. Methods Thirty male C57BL/6J mice were divided randomly into control group, model group and ginsenoside Rg1 intervention group. Myocardial tissues of each group were taken after successful modeling, and the myocardial histological changes were observed through hematoxylin-eosin staining(HE) and Masson staining(Masson). Western blot assay was used to detect the expressions of apoptotic proteins such as TLR4, MyD88, Bax and Bcl-2 in TLR4/MyD88 signaling pathway, and polymerase chain reaction(PCR) was used to detect the expressions of genes like TLR4, MyD88, Bax, and Bcl-2 in the myocardial tissue. Results HE staining and Masson staining showed that the morphology of myocardial cells in the control group was normal and the cells were arranged in order; and the morphology and arrangement of myocardial cells in the ISO group after fibrosis were irregular. Western blot results showed that, compared with the control group, the protein expressions of TLR4, MyD88 and Bax in myocardial tissue of mice in ISO group increased significantly, while the expression of Bcl-2 decreased, which was significant statistically(P<0.05). Compared with ISO group, the expressions of TLR4, MyD88 and Bax in the myocardial tissue of the mice in Rg1 group decreased significantly, while the expression of Bcl-2 increased, which was significant statistically(P<0.05). PCR results showed that, compared with the control group, mRNA levels of TLR4, MyD88,and Bax in the myocardial tissue of the mice in ISO group increased significantly, while mRNA expression of Bcl-2 decreased, which was significant statistically(P<0.05). Compared with ISO group, mRNA levels of TLR4, MyD88, and Bax in myocardial tissue of mice in Rg1 group decreased significantly, while mRNA expression of Bcl-2 increased, which was significant statistically(P<0.05). Conclusion Ginsenoside Rg1 may affect the activation of TLR4/MyD88 signaling pathway in myocardial fibrosis model mice, delay the progression of myocardial fibrosis, and play a role in cardiac protection.
