亚慢性砷染毒对大鼠精子形态的影响

摘要(Abstract):

目的:探讨亚慢性砷染毒对大鼠精子超微结构的影响。方法:清洁级SD雄性儿童期大鼠40只,体重100~120 g,随机分为对照组、低剂量组、中剂量组和高剂量组;染砷组自由饮用含三氧化二砷(As2O3)的蒸馏水,低、中、高剂量的砷浓度分别是25、50和100 mg/L,对照组自由饮蒸馏水,连续喂养3个月;实验结束后处死大鼠,采用扫描电镜(SEM)观察大鼠精子超微结构的改变。结果:对照组、低、中、高剂量组顶体完整率分别为(92.4±8.57)%、(85.7±7.7)%、(70.4±6.6)%、(50.0±6.11)%,头部畸形率分别为(8.5±3.6)%、(15±5.9)%、(34.0±9.8)%、(49.8±6.44)%,尾部畸形率分别为(10.2±5.8)%、(16.9±8.3)%、(32.7±7.0)%、(41.8±4.7)%,中、高剂量组与对照组比较,差异均有统计学意义(P<0.05)。结论:亚慢性砷染毒能导致大鼠精子超微结构发生改变。
Objective: To investigate the mechanism of influence of sub-chronic arsenic poisoning on ultra microstructure of rats' sperm. Method: Forty Sprague-Dawley( SD) rats of childhood in cleanliness grade,weighing 100 ~ 200 g each,were divided randomly into four groups: control group,low dose group,middle dose group and high dose group,10 in each group. The rats were infected with sub-chronic arsenic poisoning by feeding with AS2O3 dissolved in distilled water. The concentration of AS2O3 in low,middle and high dose group was 25 mg / L,50 mg / L,and 100 mg / L,respectively. The rats in control group drank distilled water freely. The feeding lasted three months continuously. At the end of the feeding,all the rats were killed. The scanning electron microscope was adopted to observe the changes of ultra microstructure of rats' sperm. Results: The integrity rate of acrosome in the sperms in control,low,middle and high dose groups was( 92. 4 ± 8. 57) %,( 85. 7 ± 7. 7) %,( 70. 4 ±6. 6) %,( 50. 0 ± 6. 1) % respectively. The malformation rate of head in the sperms in control,low,middle and high dose groups was( 8. 5 ± 3. 6) %,( 15 ± 5. 9) %,( 34. 0 ± 9. 8) %,( 49. 8 ±6. 44) % respectively. The malformation rate of tail in the sperms in control,low,middle and high dose groups was( 10. 2 ± 5. 8) %,( 16. 9 ± 8. 3) %,( 32. 7 ± 7. 0) %,( 41. 8 ± 4. 7) % respectively.Compared with control group,the differences were statistically significant in middle and high dose groups( P < 0. 05). Conclusion: The sub-chronic arsenic poisoning can cause ultrastructural changes of the sperm in rats.
