摘要(Abstract):

目的:观察兔左心室内膜、中层、外膜瞬时外向钾电流(Ito)和内向整流钾电流(IK1)亚单位的表达,探讨左室复极跨室壁异质性的分子机制。方法:采用Real-time PCR法测定18只健康雄性兔的左心室外膜、中层内膜瞬时外向钾电流(Ito)α亚基(Kv4.3和Kv1.4)和内向整流钾电流(IK1)α亚基(Kir2.1)的mRNA表达水平,并行统计学分析。结果:Kv4.3 mRNA在左室内膜的表达最少,与外膜、中层相比差异有统计学意义(P<0.01);Kv1.4 mRNA在左室中层的表达最少,与外膜相比有差异(P<0.05);Kir2.1 mRNA水平在左室外膜、中层、内膜的表达差异无统计学意义(P>0.05)。结论:Itoα亚基Kv4.3和Kv1.4 mRNA在左心室外膜、中层、内膜表达存在差异性,可能与左室复极跨室壁异质性有关。
Objective: To observe expression of transient outward K+current( Ito) and inward rectifier K+current( IK1) subunits mRNA in epicardium,midmyocardium and endocardium of rabbit left ventricular,and to explore the mechanism of transmural heterogeneity of repolarization in left ventricular. Methods: The mRNA expression of Itoα units( Kv4. 3,Kv1. 4) and IK1α unit( Kir2. 1) were evaluated by real-time PCR. Results: Expression of Kv4. 3 mRNA was lower in epicardium than that in midmyocardium and endocardium( P < 0. 05). Expression of Kv1. 4 mRNA was lowest in midmyocardium,and the expression difference between midmyocardium and epicardium was statistically significant( P < 0. 05). No significant difference of Kir2. 1 mRNA expression was found among endocardium,midmyocardium and epicardium. Conclusion: Kv4. 3,Kv1. 4 differently express in endocardium,midmyocardium and epicardium of rabbit left ventricular which may related to transmural heterogeneity of repolarization in left ventricular.
