肺炎Ⅰ号保留灌肠对RSV感染模型大鼠TNF-α、IL-6的干预影响

摘要(Abstract):

目的:观察肺炎Ⅰ号保留灌肠对人类呼吸道合胞病毒(RSV)肺炎模型大鼠血清中肿瘤坏死因子α(TNF-α)、白介素,6(IL-6)的影响。方法:将48只SD幼年大鼠随机分为正常对照组、空白模型组、肺炎Ⅰ号高剂量保留灌肠给药组、肺炎Ⅰ号中剂量保留灌肠给药组、肺炎Ⅰ号低剂量保留灌肠给药组及利巴韦林保留灌肠给药组,正常对照组大鼠用生理盐水、其余各组大鼠用RSV感染稀释液滴鼻3 d造模;于造模第3天开始,各组大鼠用相应试剂灌肠,连续用药干预7 d,第8天时收集各组血清、肺组织以及肺泡灌洗液,比较各组大鼠肺组织湿干比(W/D)、观察肺组织学,并采用ELISA法检测各组大鼠肺组织及肺泡灌洗液中的TNF-α、IL-6的水平。结果:肺组织病理学显示,利巴韦林、肺炎Ⅰ号干预后肺组织病变减轻,其中以中剂量组效果较为明显;肺炎Ⅰ号高、中剂量给药组、利巴韦林组W/D值较模型组明显下降,差异有统计学意义(P <0. 05);与模型组比较,肺炎Ⅰ号高、中、低剂量组、利巴韦林组各组织炎症因子水平均明显降低,差异有统计学意义(P <0. 05);与利巴韦林组比较,肺炎Ⅰ号中剂量给药组在血清、肺组织以及肺泡灌洗液TNF-α及IL-6表达含量及高剂量给药组血清TNF-α差异均无统计学意义(P> 0. 05);高、中剂量组TNF-α、IL-6水平均较低剂量组明显下降,差异有统计学意义(P <0. 05)。结论:肺炎Ⅰ保留灌肠能有效降低模型鼠RSV感染后血清、肺组织及肺泡灌洗液中TNF-α、IL-6表达含量。
Objective: To observe the effect of retention enema of Pneumonia I on TNF-a and IL-6 in serum of rats with human respiratory syncytial virus( RSV) pneumonia. Methods: Forty-eight young SD rats were randomly divided into normal control group,blank model group,pneumonia I high-dose retention enema group,pneumonia I medium-dose retention enema group,pneumonia I low-dose retention enema group and ribavirin retention enema group. Rats in the normal control group were given normal saline and the rest of the rats were given RSV infection dilution for 3 D. Rats in each group were given enema with corresponding reagents for 7 days from the 3 rd day of modeling,and serum,lung tissue and alveolar lavage fluid were collected on the 8 th day. The wet dry ratio( W/D) of lung tissue in each group was compared and lung histology was observed,the levels of TNF-and IL-6 in lung tissue and bronchoalveolar lavage fluid of rats in each group were detected by ELISA. Results: Lung histopathology showed that the lung lesions were alleviated after the intervention of ribavirin and pneumonia I,and the effect of middle dose group was more obvious. The W/D values of pneumonia No. I high and medium dose group and ribavirin group were significantly lower than those of model group( P < 0. 05).Compared with the model group,the levels of inflammatory factors in the high,middle and low dose groups of pneumonia I and ribavirin groups were significantly lower( P < 0. 05). Compared with ribavirin group,the expression levels of TNF-alpha and IL-6 in serum,lung tissue,alveolar lavage fluid and alveolar lavage fluid of pneumonia I medium dose group had no significant difference( P > 0. 05).The levels of TNF-a and IL-6 in high and middle dose groups were significantly lower than those in low dose group( P < 0. 05). Conclusion: Pneumonia I retention enema can effectively reduce the expression of TNF-a and IL-6 in serum,lung tissue and alveolar lavage fluid after RSV infection.
