血小板膜糖蛋白基因-176T/C和-506G/A单核苷酸多态性与肺癌的相关性

摘要(Abstract):

目的:探讨血小板膜糖蛋白基因(GP)-176T/C和-506G/A单核苷酸多态性与肺癌的相关性。方法:经病理确诊的肺癌患者100例作为肺癌组,同期健康体检者100例作为对照组,提取两组受检者外周全血DNA,用聚合酶链反应-序列特异性引物(PCR-SSP)法检测GP-176T/C和-506G/A位点的基因型;采用非条件logistic回归法分析肺癌患者GP-176T/C和-506G/A基因型频率及等位基因频率与肺癌的相关性。结果:对照组和肺癌组GP-176T/C基因型频率及等位基因频率比较,差异有统计学意义(P<0.05);GP-176T/C和-506G/A位点的基因型和等位基因频率分布与肺癌的TNM分期及淋巴结转移之间无明显关系(P>0.05);对GP-176T/C和-506G/A位点的不同基因型与肺癌的相关性进行相对危险度分析,结果显示GP-176T/C位点中TC型及C等位基因对肺癌的发病有统计学意义(OR=0.392、0.490,95%CI分别为0.241~0.639和0.301~0.798,P<0.05)。结论:GP-176T/C基因多态性可能与肺癌的发生有关,C等位基因可能是肺癌发病的保护因素。
Objective:To investigate the correlation between platelet membrane glycoprotein( PMGP)-176T/C and-506G/A single nucleotide polymorphisms and lung cancer. Methods:One hundred pathologically confirmed lung cancer patients were selected as observation group( group O),and 100 healthy people as control group( group C). Peripheral blood of the research subjects was collected and PMGP gene was detected for genotypes of GP-176T/C and-506G/A by using polymerase chain reaction-sequence specific primers( PCR-SSP) method. The correlation of genotype frequency and allele frequency of GP-176T/C and-506G/A with lung cancer was analyzed with non-conditional logistic regression. Results:The differences of GP-176T/C genotype and allele frequencies between groups O and C were statistically significant( P < 0. 05); The genotype and allele frequencies of GP-176T/C and-506G/A were not significantly correlated with TNM stages and lymph node metastasis( P > 0. 05); The relative risk analysis of GP-176T/C and-506G/A sites showed that at GP-176T/C locus,TC type and C allele had statistically significant influences on the incidence of lung cancer,of which OR( 95% CI) were 0. 392( 0. 241 ~ 0. 639) and 0. 490( 0. 301 ~ 0. 798)( P < 0. 05). Conclusions:GP-176T/C gene polymorphism might correlate with the occurrence of lung cancer,and C allele might be a protective factor against lung cancer.
