回转模拟失重对共培养体系血管内皮细胞Notch3信号通路相关蛋白表达的影响

摘要(Abstract):

目的:探讨模拟失重条件下人脐静脉内皮细胞(HUVECs)和人主动脉平滑肌细胞(HASMCs)共培养的细胞模型体系中HUVECs的Notch3信号通路相关蛋白表达。方法:分别取HUVECs和HASMCs,分为正常重力组和模拟失重组,共培养48 h时,观察HUVECs形态;提取两组HUVECs细胞的总蛋白,采用Western blot技术检测Notch3、Jagged1、c-Src和RBP-Jκ蛋白表达。结果:共培养48 h时,正常重力组HUVECs细胞呈现为多角形、以"铺路石"样紧密排列,模拟失重组HUVECs则多为圆形、边角变钝;与正常重力组比较,模拟失重组HUVECs中Notch通路相关蛋白Notch3、Jagged1、c-Src及RBP-Jκ的表达显著降低(P <0. 05)。结论:模拟失重可引起共培养模型中HUVECs细胞Jagged1和Notch3的蛋白表达降低,其机制可能与c-Src表达降低有关。
Objective: To investigate the effect of simulated microgravity on the expressions of Notch pathway-associated proteins of human aortic smooth muscle cells( HASMCs) co-cultured with human umbilical vein endothelial cells( HUVECs). Methods: The clinostat was employed to simulate microgravity( i. e.,weightlessness). Co-culture cells were divided into normal gravity group and simulated weightless group. The morphology of HASMCs was observed at 48 h after co-culture,and cells were collected to be extracted for total cell lysates. Western blot was used to detect the expression levels of Notch3、Jagged1、c-Src and RBP-Jκ. Results: The morphology of HASMCs in the normal gravity group exhibited the polygons which were closely arranged in a pavestone path,whereas HUVECs were mostly round and the edges were dull in simulated weightless group. The expression levels of Notch3,Jagged1,c-Src and RBP-Jκ were significantly lower in simulated weightless group than in the normal gravity group( P < 0. 05). Conclusion: Simulated microgravity decreased the expression levels of Jagged1 and Notch3 in HUVECs cells in the co-culture model,and its mechanism may be related to the decrease of c-Src expression.
