家蝇u93基因编码蛋白多克隆抗体的制备与鉴定

摘要(Abstract):

目的:制备特异性的u93基因编码蛋白的多克隆抗体。方法:采用原核表达技术,体外诱导表达u93重组蛋白;采用蛋白免疫法,以纯化的重组蛋白为抗原免疫新西兰大白兔获得多克隆抗体;用ELISA法检测多克隆抗体效价,运用Western-blot方法对u93多克隆抗体的纯度和特异性进行分析。结果:制备的兔抗u93重组蛋白多克隆抗体效价为1∶4 000,Western-blot结果显示该多克隆抗体能特异性识别u93重组蛋白。结论:成功制备u93基因编码蛋白的多克隆抗体。
Objective: To prepare the specific polyclonal antibody to u93 gene encoding protein.Methods: The expression of u93 recombinant protein was induced in vitro by the technique of prokaryotic expression. Immunization with recombinant protein was used to immunize New Zealand white rabbits to obtain polyclonal antibodies. The polyclonal antibody titers were detected by ELISA,and purity and specificity of u93 polyclonal antibody were analyzed by Western-blot. Results: The polyclonal antibody titer of rabbit anti-u93 recombinant protein was 1∶ 4 000,and Western-blot results showed that the polyclonal antibody could specifically recognize recombinant u93 protein. Conclusion: The polyclonal antibody against u93 gene encoding protein is successfully prepared.
