瘤果黑种草子对小鼠急性酒精性肝损伤的影响

摘要(Abstract):

目的:探讨瘤果黑种草子对小鼠急性酒精性肝损伤的影响。方法:50只昆明小鼠随机分为正常对照组、模型组、阳性对照组及瘤果黑种草子高、低剂量组,除正常对照组外,其余各组给予56度白酒(10 m L/kg)灌胃建立急性酒精性肝损伤模型,正常对照组给予等量生理盐水;灌胃同时,阳性对照组灌胃葵花护肝宁,瘤果黑种草子高、低剂量组分别灌胃黑种草子水提液6 g/kg或2 g/kg进行灌胃,连续10 d,灌胃期间观察小鼠的体重、精神、活动及毛色等情况;最后一次灌胃后10 h时取血检测小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)的水平,同时称取小鼠体质量和肝脏质量,计算肝指数,观察小鼠肝组织外观及肝组织病理学改变。结果:与正常对照组比较,模型组小鼠体重减轻,嗜睡,毛发光泽度下降;瘤果黑种草子组小鼠情况明显改善,小鼠毛发光滑整齐,体质量增长恢复正常,高剂量组最为明显;小鼠体质量和肝指数比较,模型组低于正常对照组,瘤果黑种草子组高于模型组(P<0.05);血清ALT及AST活性比较,模型组高于正常对照组,瘤果黑种草子组低于模型组(P<0.05);模型组小鼠肝组织外观呈黄褐色、正常对照组呈红褐色,瘤果黑种草子组介于两组之间,高剂量组更接近正常对照组;HE染色结果显示,模型组小鼠肝细胞明显气球样变性、坏死,并伴有少量炎症细胞浸润;与模型组比较,高剂量瘤果黑种草子组小鼠肝细胞的病理学改变减轻。结论:高剂量瘤果黑种草子水提液对小鼠急性酒精性肝损伤有保护作用。
Objective: To explore the effects of seeds of Uygur medicine Nigella glandulifera freyn on acute alcoholic liver damage of mice. Methods: 50 mice were randomly divided into control group,model group,the positive control group,high dose of Nigella water extract group(6 g / kg),and low dose of Nigella semen water extract group(2 g / kg). The normal group received saline of equal volume while the other groups were orally administered daily 56 degree liquor of 10 m L / kg to establish a model of acute alcoholic liver injury. When intragastric administration was going on,the positive control group received gavage of sunflower Huganning,while Nigella glandulifera freyn group were given intragastric administration of Nigella seeds water extract 6 g / kg(high dose) or 2 g / kg(low dose) for 10 d.The body weight,spirit,activity and hair color of mice were observed during the period of gavage.10 h after the last intragastric administration,serum of mice was collected to detect serum glutamic pyruvic transaminase(ALT),aspartate aminotransferase(AST) levels. At the same time,the mice body weight and liver weight were measured to calculate liver index and the liver tissue appearance and the pathological changes of liver tissue were observed. Results: Compared with the normal controlgroup,model group mice showed weight loss,sleepiness,hair gloss drop. The situation of mice in Nigella glandulifera freyn group were significantly improved. Their mouse hair was smooth,body weight increased and returned to normal with high dosage groups being better. The body weight and liver index of mice in model group were lower than their counterparts of normal control group while the body weight and liver index in Nigella glandulifera freyn group were higher than their counterparts of model group( P < 0. 05). The serum ALT and AST activity in model group were lower than their counterparts of normal control group while the serum ALT and AST activity in Nigella glandulifera freyn group were higher than their counterparts in model group( P < 0. 05). The liver tissue appearance of model group was brown while normal control group reddish brown. The appearance of Nigella glandulifera freyn group was between model group and normal control group,with the high dose group closer to the normal control group. He staining results showed that mice hepatic cells of model group showed significant ballooning degeneration and necrosis,accompanied by a small amount of inflammatory cell infiltration.Compared with model group,the pathological changes of mice liver cell of high dose of Nigella glandulifera freyn was alleviated. Conclusion: High dose of Nigella glandulifera freyn water extract has a protective effect on acute liver injury in mice.
