艾迪注射液对裸鼠人胃癌移植瘤细胞凋亡的影响

摘要(Abstract):

目的:观察艾迪注射液对裸鼠人胃癌移植瘤细胞凋亡的影响。方法:将人胃癌细胞BGC-823接种于裸鼠右侧腋皮下,建立荷瘤裸鼠模型。将荷瘤裸鼠随机分为:模型、环磷酰胺及艾迪注射液高、中、低剂量5个组,加上1个对照组(未接种肿瘤细胞),各组动物按各自药物和剂量腹腔注射(ip)给药,连续14d。末次给药后处死动物,剥取瘤块,用流式细胞仪测定移植瘤组织的细胞凋亡率,并用电子显微镜观察其超微结构。结果:艾迪注射液高剂量组的细胞凋亡率明显高于模型组,电镜下可见明显的凋亡表现。结论:艾迪注射液具有促进裸鼠人胃癌移植瘤组织细胞凋亡的作用。
Objective:To observe the effect of Aidi injection(Ad)on apoptosis of transplanted tumor of human gastric cancer in nude mice.Methods:Gastric cancer model of nude mice were established by transplanting human gastric cancer cell line(BGC-823)into hypoderm at right armpit,and then the cancer-bearing mice were divided randomly into 5 groups:model group,cyclophosphamide(CTX)group,and 3 Ad treated groups(high-dose,middle-dose,and low-dose groups).Physiologic saline(PS),CTX,Ad were given intraperitoneally to mice of different groups respectively for 14 days.A normal negative group was set up composed by nude mice without transplantation and administrated with PS(ip).The mice were sacrificed by decapitation,and the tumor tissues were extracted for observation of apoptosis degree by flow cytometry detection,and cancer cell ultrastructure by electron-microscope(EM).Results:Tumor inhibitory rate was 61.84% in Aidi high dose group of which the cell apoptosis rate was obviously higher than that of model group(P<0.05).Ultrastructure of cancer cell apoptosis was observed under EM.Conclusions:Aidi injection has effects to apparently increase the apoptosis of human gastric cancer in nude mice.
