海洛因依赖对大鼠颌下腺5-羟色胺免疫反应细胞的影响

摘要(Abstract):

目的:研究海洛因依赖期间大鼠颌下腺内5-羟色胺(5-HT)免疫反应(immunoreactive,IR)细胞表达及功能的改变,探讨颌下腺5-HT-IR细胞在海洛因依赖期间的可能作用。方法:皮下注射海洛因建立海洛因依赖大鼠模型,用免疫组织化学SABC法并结合图像分析方法进行研究。结果:海洛因依赖期间,5-HT-IR细胞的免疫染色明显增强。图像分析结果显示,5-HT-IR细胞数量增加,平均灰度值明显降低,且第24~31天时变化最明显(P<0.05)。结论:海洛因依赖期间,颌下腺5-HT-IR细胞的表达与功能均发生了改变,提示颌下腺5-HT-IR细胞可能参与调节海洛因依赖对机体的损伤过程。
Objective: To study the changes of expression and function of 5-HT immunoreactive(IR) cells in the submandibular gland of rats for investigating the possible function of 5-HT-IR cells during heroin dependence.Methods: Heroin dependence model of rats was established by subcutaneous injection of heroin.Immunohistochemical SABC method and image analysis were used in the study.Results: The immunohistochemical staining intensity of 5-HT-IR cells from heroin dependent rats increased obviously.The image analysis results showed that the number and mean grey degree of 5-HT-IR cells increased obviously,especially during the 24th-31st days of heroin dependence(P<0.05).Conclusions: The expression and function of 5-HT-IR cells change during heroin dependence,which suggests that the 5-HT-IR cells in the submandibular gland might be involved in the regulation process of heroin dependence.
