京尼平对肾癌细胞解偶联蛋白2及线粒体能量代谢的作用

摘要(Abstract):

目的:探讨京尼平对肾癌细胞株(GRC-1)的作用及对解偶联蛋白2和线粒体能量代谢的影响。方法:使用不同剂量京尼平干预GRC-1后,应用MTT法检测细胞增殖和凋亡情况;使用RT-PCR法检测UCP2 mR-NA表达水平;收集细胞,检测细胞内ATP含量;提取线粒体,使用荧光分光光度计检测吸光度的变化,间接反映线粒体膜通透性转运孔开放程度。结果:随着京尼平剂量的增高,GRC-1细胞的抑制率逐渐升高,与对照组相比,中、高剂量京尼平组细胞的抑制率明显升高(P<0.05),UCP2的表达下调,UCP2 mRNA在高剂量京尼平组中的表达明显降低(P<0.05),且中、高剂量组线粒体内ATP含量下降(P<0.05),中、高剂量组线粒体膜通透性转运孔开放程度显著提高(P<0.001)。结论:京尼平可明显抑制肾癌细胞株的增殖,降低UCP2表达水平并对线粒体能量代谢产生影响。
Objective: To investigate the effect of genipin on the apoptosis of kidney cancer cell line represented by GRC-1,UCP2,and mitochondrial energy metabolism.Methods: GRC-1 was treated by genipin at different doses,and its proliferation and apoptosis were detected by MTT assay.RT-PCR was used to assess the expression levels of UCP2 mRNA.ATP content in GRC-1 was measured,and mitochondria were extracted.Fluorospectrophotometer was adopted to test the absorbance which could reflect membrane permeability of mitochondria.Results: As comparing with those in control group,apoptosis rate of GRC-1 cells in medium and high genipin groups increased obviously(P<0.001).while UCP2 mRNA expression level reduced.ATP amounts,and mitochondrion optical density in medium and high genipin groups were significantly lower than those in control group(P<0.05).Conclusion: Genipin could inhibit GRC-1 growth obviously,of which the mechanism might be the decrease of UCP2 expression and the influence on mitochondrial energy metabolism.
