Cx43蛋白、雌激素受体、孕激素受体在子宫内膜癌组织的表达及相关性研究
Study on Expression of Connexin 43 Proteins, Estrogen Receptor and Progesterone Receptor in Endometrial Carcinoma

摘要(Abstract):

目的: 探讨细胞间隙连接蛋白(Cx43)、雌激素受体(ER)、孕激素受体(PR)在子宫内膜癌中的表达以及Cx43蛋白与ER、PR的相互关系。方法:应用SP免疫组织化学方法,检测30例子宫内膜癌组织(内膜癌组)、30例正常增生期子宫内膜组织(正常组)中的Cx43蛋白、ER及PR的表达。结果: (1)Cx43蛋白、ER及PR在正常组的表达阳性率分别为80 0%、83 3%及86 7%,在子宫内膜癌组中的表达阳性率分别为46 7%、50 0%及56 7%,两组比较差异有显著性(P<0 05); (2)Cx43蛋白、ER及PR在病理分级为G1 的内膜癌中阳性率分别为80 0%、80 0%及80 0%,高于G2 (60 0%、60 0%及46 7% )和G3 者(10 0%、20 0%及30 0% ),P<0 05; (3)浅肌层浸润的内膜癌阳性率分别为59 1%、63 6%及86 3%,高于深肌层浸润者(均为12 5% ),P<0 05; (4)手术分期为Ⅰ~Ⅱ期的内膜癌阳性率分别为54 2%、50 0%及62 5%,Ⅲ~Ⅳ期者的阳性率分别为16 7%、50 0%及33 3%,两者比较差异无显著性,P>0 05。结论:Cx43蛋白、ER及PR与内膜癌发生有关,可作为诊断、了解病变程度和监测预后的重要指标之一,Cx43蛋白与ER及PR的表达呈现一致性。
Objective:To investigate the expression of connexin 43 proteins, estrogen receptor (ER) and progesterone receptor (PR) in human endometrial carcinoma tissue and relations among them. Methods: Changes of expression of the gap junction proteins-connexin 43 (Cx43), estrogen receptor (ER) and progesterone receptor (PR) in human normal endometrium (30 cases) and endometrial cancer samples (30 cases) were determined with immunohistochemistry techniques. Results: 1) In normal endometrium tissues, the detection rates of Cx43 protein, ER and PR were respectively 80.0%, 83.3% and 86.7%; But in endometrial carcinoma positive expression of the three kinds of proteins were respectively 46.7%, 50.0% and 56.7%; The differences were significant.(P<0.05); 2) The positive expression of Cx43 proteins, ER and PR in gradeⅠcarcinoma cells were respectively 80.0%, 80.0% and 80.0%, which were higher than those in grade Ⅱ(60.0%,60.0% and 66.7%) and grade Ⅲ (10%,20.0% and 30.0%); 3) They were also higher in endometrial carcinoma with superficial infiltration (59.1%, 63.6% and 72.7%) than in those with deep infiltration (12.5%,12.5% and 12.5%, P<0.05);4) Their detection rates in carcinoma cells of operation stages Ⅰ-Ⅱ were respectively 54.2%, 50.0% and 62.5%, while they were respectively 16.7%, 50.0% and 33.3% in carcinoma cells of operation stages Ⅲ-Ⅳ. The differences were not significant (P>0.05); Conclusions: Cx43 proteins, ER and PR expression are significantly associated with pathological grade and depth of myometrial infiltration(P<0.05), and could be one of the indexes for diagnosis of endometrial carcinoma, and for the monitoring of the development and prognosis of the disease. The expression level of Cx43 accords well with the expression of ER and PR.
