明胶海绵与人胚半月板细胞构建复合物的研究A Study on Construction Complex of Human Fetal Meniscus Cells and Gelatin Sponge

摘要(Abstract):
目的:研究明胶海绵作为半月板组织工程支架材料应用的的可行性。方法:采用胶原酶阶段消化 法获取人胚半月板纤维软骨细胞,取经胰岛素样生长因子I(IGF Ⅰ)扩增的第3代细胞与已用多聚赖氨酸修饰 的明胶海绵复合培养,进行MTT、光镜、扫描电镜观察。结果:纤维软骨细胞于明胶海绵上牢固粘附,4d开始增 殖,3周时已有大量细胞生长于明胶海绵上。结论:明胶海绵表现出良好的亲水性、细胞吸附性和体外生物相容 性,适于作为组织工程细胞培养支架材料用。
Objective: To investigate the possibility of gelatin sponge as fibrocartilage cell scaffold in meniscus tissue-engineering. Methods: Human fetal meniscal fibrochondrocytes were collected with collagenase digestion method. The 3th passage cells proliferated with IGF-I were co-cultured with gelatin-sponge that modified by poly-Lysine and were observed with MTT, light microscop and scanning electronic microscopy. Results: Fibrocartilage cells attached to gelatin sponge closely and begin to proliferate in 4 days. lots of cells were found to proliferate onto gelatin sponge in 3 weeks. Conclusions: Gelatin sponge has good hydrophilia, adsorptivity, biocompatibility with fibrocartilage cells. It can be used as scaffold for meniscus tissue engineering.
