TGF-β_1与IGF-Ⅰ对体外培养兔软骨细胞增殖的影响

摘要(Abstract):

目的:探讨转化生长因子 β1(TGF β1)与胰岛素样生长因子Ⅰ(IGF Ⅰ)联合作用对关节软骨细胞增 殖行为的影响。方法:采用兔关节软骨细胞体外单层培养方法,将TGF β1与IGF Ⅰ作用于软骨细胞,并用MTT 比色分析法对其增殖行为进行分析,探讨多因子对关节软骨细胞增殖行为的影响。结果:(1)TGF β1组的最佳 效应浓度为5μg/L,IGF Ⅰ组的最佳效应浓度为50μg/L。(2)TGF β1组(5μg/L)、IGF Ⅰ组(50μg/L)以及两 因子联合使用组软骨细胞增殖均较对照组高(P<0.01),联合使用组促增殖作用最强,优于单一的TGF β1最佳 效应浓度组和IGF Ⅰ最佳效应浓度组(P<0.05)。结论:TGF β1与IGF Ⅰ联合运用早期促软骨细胞增殖作用 优于单一的TGF β1或IGF Ⅰ,在软骨细胞增殖过程中TGF β1和IGF Ⅰ有良好的协同作用。
Objective: To investigate the combined effects of transforming growth factor-β 1 (TGF-β 1) and insulin-like growth factorⅠ(IGF-Ⅰ) on rabbit articulation cartilage cell proliferation. Methods: Rabbit articulation cartilage cells were cultured in vitro. The second generation cells were treated by DMEM containing different growth factors. Dose-response were observed by MTT colorimetric method. Results: 1) The optimal concentration of TGF-β 1 was 5.0μg/L, and that of IGF-Ⅰwas 50.0μg/L. 2) Both the growth factors were potent mitogens. They had mitogenic effect and acted dose-dependently when was used singly. The effects were significant in 72 hours. Among them, the effect of TGF-β 1/ IGF-Ⅰ composite was the best. The synergetic effects were achieved when they were used in combination. Conclusions: These data demonstrate that TGF-β 1/ IGF-Ⅰ composite has better promoting effect than single TGF-β 1 or IGF-Ⅰ on cartilage repair. It is suggested that optimal combination of TGF-β 1/ IGF-Ⅰcan promote the proliferation of rabbit articulation cartilage cells significantly.
