吡喹酮驱虫治疗前后牛肉绦虫形态和组织化学的研究

摘要(Abstract):

<正> 牛肉绦虫(Jaenia saginata Goeze,1782)的形态、生活史、流行病学等久已为人所熟知;但其组织结构和组织化学方面除见有Chowdhury et al,(1955,1956,1962);Logachev,(1953)等人的零星报道外,尚未见到系统的研究。吡喹酮(Praziquantel)是近年来国内外广泛报道的一种广谱抗寄生虫新药,它对于驱除人、畜多种寄生扁形动物均有良效。因此研究吡喹酮的驱虫作用机制对于该药的进一步推广、提高以及新药的寻求都具有重要的理论和实际意义。这方面已有杨元清等(1979),肖树华等(1980)和沈一平(1981)以及Pax & Fetterer(1979);Andrews
1. Through observations on specimens taken from gastroenteric operations, it was discovered that the histological structures and histochemical components of Taenia saginata Goeze, 1782 were similar to other cyclophyllidean cestodes. The majority of glycogen were found in parenchyma, muscles, vitelline cells and eggs but absent in most reproductive organs. The distribution of AKP was observed in tegument, tegumental cells, reproductive cells and the wall of uterus and excretory ducts; but ACP was only in tegument. Nucleic acid was more abundant in embryo-cells, reproductive cells and tegumental cells. There were strong reactions of basic protein in the tegument, tegumental cells, vari- ous muscles and vitelline globes etc. 2. After treatment with a single dose of praziqantel at 10 mg/kg, the tapeworms were discharged within 3 to 24 hours. These worms were found immobilized and greatly contracted when expelled. The histological changes observd undeer microscope consisted mainly contractions of muscles and sper- matozoa; wrinkling, vacuolization, swelling and breaking in tegument, subtegument and parenchymal organs. These changes finally led to the dis- ruption of structures of the whole worm. After treatment with cucurbita seed-areca nut, the worms were expelled within 6 hours. These worms were yet alive and mobile, but it was obser- ved that the tegumental cells of scolex and neck regions became round and their cytoplasma disappeared. 3. The histochemical changes resulted from praziquantel treatment were observed as follows: (1) The glycogen granules within parenchyma disin- tegrated or even disappeared; (2) The activities of AKP and ACP of tegument markedly decreased; (3) The granules of positive reaction of DNA difused and weakened but the reactions of RNA within reproductive organs had no appearent changes or had only a little diminished within 5 hours after treat- ment; (4) on the contrary, the reactions shown by mercury-bromophenal blue method for basic protein in muscles were stronger than those in the control. Cucurbita seed-areca nut caused disintegrating and decreasing of glycogen gra- nules; disappearing of the positive reactions of AKP and ACP. But there were no changes on the reactions of DNA, RNA and basic protein. 4. It was suggested that the results of strong contraction of muscles and dis- ruption in tegument caused by praziquantel are: exhaustion of energy, inter- ference of the absorption of nutrient, quick resolving of substances of energy and disordering of intermediate metabolism processes. Besides, the organic substances such as glucose and α-amino acid etc. leaked out through the da- maged tegument and the digestive fluid of the host penetrated into the worm. All of these changes made the tapeworms collapsed, dead and expelled. Cucurbita seed-areca nut couldn't directly, kill the worm but interfering the absorption of nutrients and might made the worm paralysed temporarily, thus expelled by the host.
