依脉特灵对ca_-~(2+)所致小鼠离体子宫平滑肌收缩的拮抗作用

摘要(Abstract):

本文观察到IT对小鼠离体子宫平滑肌高K~+去极化后Ca~(2+)所致的收缩有显著抑制作用。CaCl_2量效曲线IT对其呈非竞争性拮抗。在无Ca~(2+)高K~+溶液中,IT10μM抑制OXY依赖细胞内Ca~(2+)引起的收缩,而IT50μM对依赖细胞内外Ca~(2+)所致子宫肌收缩均有抑制作用。Ver对小鼠离体子宫的作用,远较IT的强。但Ver较高浓度(59nM)只抑制依赖细胞内Ca~(2+)的收缩,对依赖细胞外Ca~(2+)的收缩影响甚少。IT与Vcr拮抗ca~(2+)的作用相似。IT为Ca~(2+)拮抗剂。
Isotetrandrine (IT) antacalcium on contraction of isolated mouse uterus were investigated. The isolated mouse uterus contraction which induced by Ca~(2+) (0.2mM) after high K~+-depolarization was markedly relaxed by IT 50uM and verapamil (Ver) 50nM. The percentages of inhibition were 66.0% and 60.0% respectively. In Re-incubation for 30min, IT 10μM, 50μM and Ver 50nM indued dosedependent rightward displacements of the concentration-response curves for Ca~(2+)(0.01~2.0mM)and the maximal responses to Ca~(2+) were depressed, showing a non-competitive antagonism. The PD_2' values of IT and Ver were 5.03 and 7.52 respectively. The effect of Ver was stronger than that of IT. The oxytocin Ca~(2+)-dependent contraction (it occured in high K~+-Ca~(2+) free depolarized solution) and Ca_0-dependent comtration (a futher contraction after the Ca~2-concentration in the both solutions was restored). The Ca-dependent contraction was depressed obviously by IT 10μM, 50μM and Ver 50nM but the Ca_0-dependent was inhibited only IT 50uM. These results indicate that IT could be a Ca~(2)-antagonist very simillar to Ver on isolated mouse uteri.
