糖尿病大鼠肾间质TGF-β_1和α-SMA的表达与细胞外基质沉积的关系

摘要(Abstract):

目的:观察糖尿病大鼠肾小管间质中转化生长因子(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)表达与纤维连接蛋白(FN)和胶原Ⅲ型(ColⅢ)沉积的关系。方法:大鼠随机分为正常对照组(A组),糖尿病1周组(B组),糖尿病2周组(C组),糖尿病1月组(D组)和糖尿病2月组(E组)。用免疫组化方法检测肾间质TGF-β_1、α-SMA、FN和ColⅢ,PAS染色观察肾组织形态学改变,生化法测定大鼠血糖、血脂、血尿肌酐以及尿蛋白。结果:糖尿病2周开始肾小管见TGF-β_1表达,并随病程进展而逐渐增多。糖尿病1月肾间质中见FN和ColⅢ沉积开始增多,2月时更多,且可见肾间质中有α-SMA表达;FN和ColⅢ增多与TGF-β_1、α-SMA呈正相关。结论:TGF-β_1刺激α-SMA的表达,从而促进糖尿病大鼠肾小管间质FN和ColⅢ的沉积。
Objective:To study the relationship of expression of transforming growth factor (TGF-β1 ) and α-smooth muscle actin (α-SMA) , with the sedimentation of FN and Col Ⅲ in renal interstitium of diabetic rats. Methods: 40 rats were randomly divided into 5 groups (8 in each) . 4 of them were experimental groups of which the rats were given 6% tetraoxypyrimidine intraperitoneally to establish animal models of diabetes. Rats in control group (group A) were injected with buffer of citric acid-sodium citrate intraperitoneally instead. The diabetes animal models were executed in 1 week (group B) , 2 weeks (group C) , 1 month (group D) and 2 months (group E) respectively. TGF-β1, α-SMA,FN and ColⅢ in renal interstitium were checked with immunohistochemistry method, and the morphology of renal interstitium was examined with PAS stained specimen. Blood glucose, fats, Scr, Ucr and u-rine protein were examined with biochemical methods. Results: The expression of TGF-β, was found in the tubulo - interstitium of diabetic rats of group C, and it increased progressively along with the extension of the disease course. The deposition of FN and Col Ⅲ appeared in the renal interstitium of rats in group D and got more in rats of group E of which the expression of a-SMA was observed and positive correlations between FN, Col Ⅲ, α-SMA and TGF-β1 were found. Conclusion: TGF-β1 might stimulate the expression of α-SMA of interstitial cells and therefore enhanced the deposition of FN and Col Ⅲ.
