血浆sFas/Apo-1和Bcl-2在卵巢癌的表达水平及意义

摘要(Abstract):

目的 :研究凋亡相关基因sFas/Apo - 1和Bcl- 2血浆中的表达水平 ,探讨二者在卵巢癌发生发展中的作用及临床应用价值。方法 :采用ELISA法对 2 0例上皮性卵巢癌、19例卵巢良性肿瘤、18例正常人群血浆中sFas/Apo - 1和Bcl- 2水平进行检测。结果 :卵巢癌患者血浆sFas/Apo - 1是 (10 2 5 6 5 2± 1 5 73)ng/L ,明显高于卵巢良性肿瘤组 (6 86 12 0± 1 36 1)ng/L ,和正常人群组 (6 38 2 6 4± 1 2 4 1)ng/L ,差异有极显著意义 (P <0 0 1)。而卵巢良性肿瘤组与正常人群组间差异无显著意义 (P >0 0 5 )。卵巢癌患者血浆Bcl- 2是 (179 14 3±2 6 84 )U/ml,也明显高于卵巢良性肿瘤组 (82 4 71± 1 94 3)U/ml,和正常人群组 (6 6 4 2 0± 1 4 0 1U/ml) ,差异有极显著意义 (P <0 0 1)。卵巢囊肿组与正常人群组间差异也无显著意义 (P >0 0 5 )。血浆sFas/Apo - 1水平诊断卵巢癌的敏感性为 35 % ,特异性为 88 89%。血浆Bcl - 2水平诊断卵巢癌的敏感性为 5 5 % ,特异性为84 6 1%。结论 :Fas/Apo - 1和Bcl- 2基因通过细胞凋亡的调控共同参与了卵巢癌的发生发展 ,对二者的血清学检测有助于卵巢癌的诊治。
Objective:To investigate the role of sFas/Apo-1 and Bcl-2 in plasma in occurrence and progression of ovarian cacinoma and its predictive value.Methods: A prospective double-blinded study was performed.The expression of sFas/Apo-1 and Bcl-2 levels in 20 cases of ovarian cacinoma,19 cases of ovarian cystis and 18 cases of normol wormen were examined with enzyme linked immunoabsorbent assay(ELISA).Results:The expression level of sFas/Apo-1 in ovarian cacinoma(1025 652±1 573 pg/ml)was significantly higher than those in ovarian cystis(686 120±1 361 pg/ml)and in normol(638 264±1 241 pg/ml),P<0 01.Then the expression level of Bcl-2 in ovarian cacinoma(179 143±2 684 Iu/ml)was significantly higher than those in ovarian cystis(82 471±1 943 Iu/ml)and in normol(66 420±1 401 Iu/ml),P<0 01.At the cut point of x±s,the sensitivity and specificity of plasma sFas/Apo-1 on predicting the ovarian cacinoma were 35%,88 89%,respectively.The sensitivity and specificity of plasma Bcl-2 on predicting the ovarian cacinoma were 55%,84 61%,respectively.Conclusion: Both Fas/Apo-1 and Bcl-2 might take part in the occurrence and progression of ovarian cacinoma through regulate apoptosis The plasma sFas/Apo-1 and Bcl-2 levels may be an earlier predictor of ovarian cacinoma,and may be helpful to select chemiotherapy plan.
