疾痢杆菌株D_(15)耐药质粒DNA在大肠杆菌中的转化表达

摘要(Abstract):

本文研究耐药质粒DNA在痢疾杆菌和大肠杆菌间的转移,痢疾杆菌D_(15)具有对链霉素、氯霉素和四环素抗性,接合传递试验证实为可传递性质粒。从痢疾杆菌D_(15)提取质粒DNA,用琼脂糖凝胶电泳检测,并把它导入大肠杆菌E、Coil K_(12) W_(1485)。从而使后者表达对链霉素抗性。经多次反复试验,其转化效率在9个转化体左右。转化频率为23×10~(-7)。
Metastasis of drug-resistant plasmid DNA between Shigella and E.Coli was studied with a conduction test.It is well know that Shigella D_(15) possesses counteraction on streptomycin, chloromycetin and tetracycline.Our study indicated that the plasmid DNA of Shigella DNA abstracted from Shigella was determined with agarose gel electrophoresis, and conducted into E.Coli K_(12) W_(1485), which expressed antagonism on streptomycin.The transformation efficiency of the plasmid DNA of Shigella D15 is 9 transformants in 1mg DNA.Its transformation frequency is 23×10~(-7).
