灵芝孢子粉抑制人肝癌细胞生长的实验研究

摘要(Abstract):

目的:研究灵芝孢子粉在体内、体外对人肝癌细胞生长的抑制作用。方法:运用噻唑蓝(MTT)法研究灵芝孢子粉水溶性成分在体外对HepG2细胞的抑制作用,并利用裸小鼠的移植瘤模型研究灵芝孢子粉在体内对HepG2细胞的生长抑制作用。HE染色观察肝癌组织结构和细胞形态改变。结果:灵芝孢子粉对HepG2细胞的IC50值随作用时间增加而减小,72 h达到最低,IC50值为2.14 g/L。灵芝孢子粉为2 g/kg时,对裸小鼠移植瘤的抑制率为57.0%。镜下观察灵芝孢子粉治疗组肿瘤坏死组织较多,细胞异型性较小。结论:高浓度及高剂量的灵芝孢子粉具有抑制肿瘤细胞生长的作用。
Objective: To evaluate the inhibitory effects of Lucid ganoderma spore on Hepatocarcinoma cell strain,HepG2,both in vitro and in vivo.Methods: The Inhibitory effects of Lucid ganoderma spore were detected by MTT method in vitro and observed through in vivo studies with human tumor nude mice xenografts.The morphology of tumor cells was observed with light microscope(HE staining).Results: The results of MTT showed that the IC_(50) values decreased with the increase of time.The minimum IC_(50) value(2.14 g/L of the Lucid ganoderma spore) was obtained in 72 h.In vivo study revealed that Lucid ganoderma spore had certain experimentally therapeutic action on Hepatocarcinoma cell strain HepG2(dose:2 g/kg,inhibitory rate:57.0%).The morphology of HepG2 cells in Lucid ganoderma spore treated group showed smaller necrosis and lower degree of atypia.Conclusion: Lucid ganoderma spore could play inhibitory action on Hepatocarcinoma cell strain HepG2 at high concentration and high dosage.
