大鼠胚胎全期缺碘对中枢神经系统发育的影响

摘要(Abstract):

<正> 地方克汀病的病因和发病机制至今尚未完全阐明,一般认为是由于胚胎期缺碘、甲状腺激素不足使中枢神经系统、骨骼系统等发育障碍的结果,但在实验室中用缺碘方法复制地方克汀病动物模型还未获完全成功。我们曾在大鼠中进行实验,给已怀孕的母鼠饲以低碘饮食、并于仔鼠出生后仍继续饲以低碘饮食,观察到胚胎发育后期及新生期缺碘对出生30天后仔鼠的中枢神经系统功能和形态均有一定影响,但不十分严重。本研究在原有基础上进行实验,目的在于阐明整个胚胎期缺碘对脑发育的影响。材料和方法一、动物分组及处理采用同系Wistar大鼠,按过去胚胎期和哺乳期缺碘实验的条件进行。分两
In order to study the effects of feto-neonatal iodine deficiency on the development of the brain, female and male Wistar rats were maintained on low iodine diet (LID) for 3 months prior to mating and over the duration of pre g- nancy and lactation. After 3 months on LID. the average thyroid weight of experimental animals was 7.3 times that of the control animals received iodine- supplemented water. The serum T_4 in the experimental and control animals was 2.80±0.14μg/100ml and 7. 32±1. 55μg/100ml (P<0. 0005) The 30-40 days old pups of experimental group showed the following func- tional and morphologic abnormalities: reduced weight of the brain, decreased ability to establish stable conditional reflex, declined speed of the reaction to the electric stimulus and conditional signal, increased density of neurons and higher percentage of undifferentiated neuroblasts and less frequently branched dendri- tes of pyramidal cells in the cerebral cortex, less frequently branched dendrites of Purkinje's cells in the cerebellar cortex, and reduced myelination of nervous fibers in the radiation of cerebral corpus callosum. The thyroid of the experi- mental pups enlarged markedly, and the serum T_4 decreased significantly~ As compared with the data obtained previously from the pups of the dams fed on LID since the mid-gestation, the pups from the dams fed on LID since 3 months prior to mating showed more severe persistent impairment of the brain development.
