依那普利和丹芪复方对糖尿病大鼠肾小球PKC_α表达的影响
Effects of Enalapril and Dan-qi Compound on Expression of PKC_α in Glomeruli of Diabetic Rats

摘要(Abstract):

目的 :观察依那普利和中药丹芪复方对糖尿病大鼠肾小球蛋白激酶Cα(PKCα)、转化生长因子 β1(TGFβ1 )和纤维连接蛋白 (FN)沉积的影响。方法 :大鼠随机分为正常对照组 (A组 )和糖尿病组 (B、C、D组 )。用四氧嘧啶复制糖尿病模型 ;B组为糖尿病对照组 ;C组为糖尿病依那普利组 ,按 2mg/(kg·d)给药 ;D组为糖尿病中药组 ,按 1 8g/(kg·d)中药浸膏给药 ,持续 6 0d。用免疫组化方法检测PKCα、TGFβ1 和FN在肾小球的表达 ;用生化方法测定大鼠血糖、血脂、血肌酐和尿蛋白 ;光镜观察肾小球形态。结果 :糖尿病各组肾小球PKCα,TGFβ1 和FN的表达明显高于正常对照组 ,但用药组 (C、D组 )的表达则低于未用药组 (B组 )。形态观察显示 ,C、D组肾小球病变比B组轻。B组血糖与C、D组无明显差异 ,但血脂、血肌酐和尿蛋白则高于C、D组。结论 :依那普利和丹芪复方可抑制糖尿病大鼠肾小球PKCα 的表达 ,从而改善肾小球的形态和功能
Objective: To investigate the effect of enalapril and Dan qi compound (a compound of Chinese medicine) on the expression of protein kinase C α (PKC α) in glomeruli of diabetic rats. Methods: Rats were randomly divided into normal control group (group A) and experimental groups (group B, C, D). Alloxan was given to rats in groups B, C, and D to establish diabetic models. Rats in group C were treated with enalapril (2 mg/kg·d), and rats in group D were treated with Dan qi compound (1 8 g/kg·d), while group B did not receive any treatment. Immunohistochemistry was used to detect the expression of PKC α, TGF β 1 and FN. The morphological changes of glomeruli were checked through microscopy. Blood glucose, triglyceride, cholesterol, creatinine and urine protein were analyzed with chemical methods. Results: The expression of PKC α, TGF β 1 and FN in glomeruli of diabetic rats increased comparing with those in normal control group. However their expressions in groups C and D were lower than in group B. The pathological changes in groups C and D were slighter than in group B. The blood triglyceride, cholesterol, creatinine and urine protein in groups C and D were lower than those in group B. Conclusion: The results suggest that enalapril and Dan qi compound may inhibit the expression of PKC α in glomeruli of diabetic rats and therefore improve glomerular structure and function.
