吡啰红G分光光度法测微量肝素
Spectrophotometric Determination of Trace Heparin with Pyronine G

摘要(Abstract):

目的:研究肝素对吡啰红G的褪色反应,建立一个简单、灵敏、选择性高的肝素测定方法。方法:分光光度法。结果:在pH3.78~6.09的B ritton-Rob inson缓冲溶液中,肝素与吡啰红G形成离子缔合物,染料发生明显的褪色,体系吸光度的降低与肝素钠浓度成正比,肝素浓度在0~2 mg/L范围内符合比耳定律,分光光度法具有较高的灵敏度,摩尔吸光系数为1.65×106L.mol-1.cm-1;共存物质的影响研究表明,分光光度法选择性好,用于肝素钠注射液效价的测定,结果满意。结论:分光光度法可用于低剂量和超低剂量肝素的定量分析。
Objective: To find a simple,sensitive,and selective method for the determination of trace heparin through the comparative observation on the discoloration effect of heparin to Pyronine G Methods: Spectrophotometry was adopted.The discoloration reaction between heparin and Pyronine G was in the Britton-Robinson buffer solution at pH3.78~6.09.Results: The absorbance reduction was directly proportional to the concentration of heparin.The linear range of heparin was from 0 to 2 mg/L.The molar absorption coefficient,ε,was 1.65×10~6 L ·mol~(-1)·cm~(-1).The study on effects of coexisting substances showed that the method had good selective function and satisfactory results had been obtained when applied to determine the potency of heparin sodium injection.Conclusion: This method could be applied to the determination of trace heparin.
