卷柏总黄酮对支气管哮喘大鼠气道的影响

摘要(Abstract):

目的:观察卷柏总黄酮(TFST)对哮喘大鼠气道炎症的影响,并探讨其可能机制。方法:将48只雄性SD大鼠随机分为对照组、哮喘模型组、TFST组,哮喘组与TFST组腹腔注射及雾化吸入卵蛋白诱导大鼠哮喘发作,TFST组预先给予TFST干预,对照组给予生理盐水;酶联免疫吸附法(ELISA)检测各组肺泡灌洗液(BALF)中白细胞介素-5(IL-5)水平,并进行炎性细胞计数,SABC免疫组织化学法检测气道上皮细胞信号转录因子与转录活化因子1(STAT1)的表达,并对各指标的关系进行相关性分析。结果:TFST组大鼠气道炎症病理改变较哮喘组减轻,BALF中IL-5水平、白细胞总数(WBC)、嗜酸性粒细胞数(EOS)及支气管上皮细胞STAT1表达水平低于哮喘组(P<0.01);BALF中IL-5水平与气管上皮细胞STAT1水平呈正相关(r=0.56,P<0.05)。结论:TFST可降低哮喘大鼠IL-5的表达和炎性细胞聚积,与抑制STAT1的表达有关。
Objective: To investigate the total flavonoids of Selaginella tamariscinaon( TFST) on airway inflammation and the expression of STAT1 protein in bronchial epithelium of asthma rats. Methods: 48 male rats were randomly divided into control group,bronchial asthma model group and TFST group. The asthma model of rats was set up with nebulization aspiration and intraperitoneal injection of ovalbumin( OVA). The TFST group was given TFST protection interventions in advance. Using ELISA for each group of bronchoalveolar lavage fluid( BALF) of interleukin-5( IL-5) concentrations were measured and to detect the number of inflammatory cells in BALF. STAT1 protein expression in airway epithelial cells was determined by SABC immunohistochemistry method. Results: The pathological changes of airway inflammation in TFST group were significantly reduced,of which level of IL-5,WBC counts and EOS counts in BALF were significantly decreased compared with that in the asthma model group( P < 0. 01). The STAT1 protein expression in bronchial epithelium and the concentration of IL-5in BALF were obviously increased in bronchial asthma model group,and positively related. The expression of STAT1 in TFST group was significantly lower than those in asthma model group. Conclusion:TFST can inhibit the IL-5 of asthmatic rats and accumulation of inflammatory cells,the mechanism may involve in inhibiting the expression of STAT1.
