吉祥草水提物对小鼠慢性阻塞性肺疾病的作用和机制

摘要(Abstract):

目的:探讨吉祥草水提物对小鼠慢性阻塞性肺疾病(COPD)的作用及机制。方法:采用随机数字表法将昆明种小鼠其分为正常对照组(n=20)、模型组(n=30)和治疗组(n=20),模型组与治疗组小鼠通过过滤嘴香烟烟熏法建立COPD模型,治疗组在过滤嘴香烟烟熏8周后,予小鼠吉祥草水提物灌胃治疗15 d,模型组和正常对照组予以同剂量纯水灌胃;于灌胃15 d后,采用Flexi Vent肺功能仪检测小鼠气道阻力(Rrs)、气道弹性阻力(Ers)、动态顺应性(Crs)、主气道阻力(P-3Rn),ELISA法检测血清中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、环氧合酶-2(COX-2)、前列腺素E2(PGE2)和基质金属蛋白酶-9(MMP9)等炎症相关因子的含量,肺组织HE染色观察小鼠肺组织病理学变化。结果:与正常对照组比较,模型组小鼠呼吸系统Rrs增加、Ers增加、Crs降低以及P-3Rn增加(P <0. 05或P <0. 01),且血清中IL-1β、IL-6、COX-2、PGE2和MMP9升高(P <0. 05),说明小鼠COPD模型造模成功;与模型组比较,治疗组小鼠Rrs降低、Ers降低、Crs升高以及P-3Rn降低,并且血清中IL-1β、IL-6、COX-2、PGE2以及MMP9表达水平降低,差异有统计学意义(P <0. 05);肺脏HE染色显示,与模型组比较,治疗组小鼠气道结构未见明显异常,炎性细胞浸润减少,肺泡结构更完整,大小趋于正常。结论:吉祥草水提物可有效治疗烟熏所致的小鼠COPD症状,其机制可能与抑制炎症因子的表达、修复COPD小鼠肺脏病理损害有关。
Objective: To investigate the effect of fluid extract from herba euphorbiae on mice with chronic obstructive pulmonary disease( COPD) and its mechanism. Method: Seventy Kunming mice were randomly divided into control( n = 20),model( n = 30) and treatment groups( n = 20). The mice in the model and the treatment groups were smoked using filtered cigarettes for 8 weeks to establish COPD model. The treatment group was given with fluid extract from Herba Euphorbiae by gavage for 15 days,and the model group and the normal control group were given the same dose of pure water by gavage for 15 days. After 15-day treatment,the FlexiVent pulmonary function meter was used to detect mouse airway resistance( Rrs),airway elastic resistance( Ers),dynamic compliance( Crs),main airway resistance( P-3 Rn). Serum interleukins 1β( IL-1β),IL-6,cyclooxygenase-2( COX-2),prostaglandin E2( PGE2) and matrix metalloproteinase-9( MMP9) and other inflammationrelated factors were detected using their corresponding ELISA assay kits. The lung tissues were stained with H&E to observe the pathological changes. Results: Compared with the control group,Rrs,Ers,and P-3 Rn were increased,while Crs was decreased in the model group( P < 0. 05 or P < 0. 01).Meanwhile,the serum levels of IL-1β,IL-6,COX-2,PGE2,and MMP9 were significantly increased( P < 0. 05),indicating that the mouse COPD model was successfully established. Compared with the model group,Rrs,Ers,and P-3 Rn were downregulated and Crs was upregulated in the treatment group. Additionally,the levels of serum IL-1β,IL-6,COX-2,PGE2,and MMP9 were decreased in the treatment group relative to control group( P < 0. 05). HE staining unveiled that when compared to control group,the airway structure was not obviously abnormal,and inflammatory cell infiltration was reduced. Moreover,alveolar structure is complete,and its sizes were close to normal sizes in the treatment group. Conclusion: Fluid extract from herba euphorbiae can effectively reduce the symptoms of mice with COPD. Its mechanism may be related to inhibiting the expression of inflammatory factors,thus repairing lung pathological damage in mice with COPD.
