大乌泡水提物的镇痛作用研究

摘要(Abstract):

目的:研究大乌泡水提物对小鼠的镇痛作用及血清中NO含量的影响。方法:热板致痛法及冰醋酸扭体法两种方法下各选择50只小鼠,每种方法下的小鼠均分为5组,分别为大乌泡根水提物高剂量组(5.2 g/kg生药)、中剂量组(2.6 g/kg生药)、低剂量组(1.3 g/kg生药)、阳性组(2.0 mg/kg盐酸吗啡)以及阴性组(生理盐水);热板致痛法观察小鼠的热板痛阈值和血清一氧化氮(NO)含量变化,冰醋酸扭体法观察小鼠扭体发作潜伏期、扭体次数、镇痛抑制率及血清NO水平。结果:与阴性组比较,热板法中阳性组小鼠给药后痛阈值升高,血清NO值降低,扭体法中小鼠扭体发作潜伏期升高,扭体次数减少,镇痛抑制率增高,差异均有统计学意义(P<0.05),说明热板致痛法和冰醋酸扭体法中小鼠模型成功;热板法中,与阴性组比较,大乌泡水提物高、中、低剂量组小鼠痛阈值升高,血清NO含量降低,差异有统计学意(P<0.05);扭体法中,与阴性组比较,大乌泡根水提物高、中剂量组小鼠扭体发作潜伏期升高,高、中、低剂量组10 min内扭体次数减少、镇痛抑制率增加,高、中剂量组小鼠血清中NO含量降低,差异有统计学意义(P<0.05)。结论:大乌泡水提物具有镇痛作用,其作用机制可能与降低小鼠血清中NO含量有关。
Objective: To investigate the analgesic effect of the water extract of Rubus multibracteatus Levl. et Vant and observe the level of the nitric oxide content in the rat serum. Methods: Acetic acid writhing method and Hot-plate method were used to study the analgesic activities. Under the two methods,the mice were randomly divided into 5 groups: high dose group(5. 2 g / kg crude drug),middle dose group(2. 6 g / kg crude drug),low dose group(1. 3 g / kg crude drug),positive control group(2. 0 mg /kg morphine hydrochloride) and negative control group(normal saline). Hot plate induced pain threshold and changes of serum nitric oxide(NO) content in mice were observed under Hot-plate method while the latent period,the times of twisting,the inhibition rate of analgesia and the level of serum NO in mice were observed under Acetic acid twisting method. Results: Compared with the negative control group,the pain threshold was increased after treatment in positive group and NO level was decreased under Hot-plate method while the writhing body seizure latency was increased,writhing times were reduced and analgesic inhibition rate was increased under Acetic acid writhing method. The differences were statistically significant(P < 0. 05),which showed that the mouse model of Hot plate method and Acetic acid writhing were successfully established. In Hot plate method,compared with the negative control group,in high,middle and low dose group,the mice pain threshold was increased,serum NO content was reduced,and the difference was statistically significant(P < 0. 05).In Acetic acid writhing method,compared with the negative control group,the writhing body seizure latency was increased in high and low dose group,writhing times were reduced and analgesic inhibition rate was increased in high,middle and low dose group,and the difference was statistically significant(P < 0. 05). Conclusion: The water extract of Rubus multibracteatus Levl. et Vant shows significant analgesic effect,whose mechanism may be related to its power to reduce NO conten in serum.
