两种成色反应的比较和玻璃纸平板包埋法的应用
A Comparative Study of Two Chromogenie Reaction and the Use of Glass Paper Flat-Embedding Method

摘要(Abstract):

本文分别用SNP和AHM作TMB反应的稳定剂,对树鼩脊髓前角运动神经元作了比较观察,结果发现两种成色反应对显示神经元胞体和近端树突无明显差别,对显示远端树突,TMB-SNP反应明显优子TMB-AHM反应。同时,本文采用的玻璃纸平板包埋法较Aldes法简便,且效果相同。
The morphology of the spinal anterior horn motoneurons in the tree shrew was comparatively studied under light microscope by using of SNP (sodium nitrop- russide, SNP) and AHM (ammonium heptamolybdate, AHM) as the stablizer in TMB(tetramethyl benzidine, TMB)chromogenic reactions of the CB-HRP method respectively. Then, the sections reacted by TMB-AHM were embedded with the technique of combined flat-embedding of glass paper(the modified Aldes' combi- ned flat-embedding technique)for electron microscopic observation, It was found that: both TMB-SNP and TMB-AHM reactions shrow the features of the neuron bodies and their proximal dendrites similarly; while in the features of the distal dendrites, especially the dendrites in the white matter, TMB-SNP reaction was more excellent than TMB-AHM On the other hand, under electron microscope, the ultrastructures(synapse, neurofibriles, mitochondria, etc. )were clearly seen in the samples of TMB-AHM reaction, but foggily in TMB-SNP reaction. The procedure of glass paper flat-embedding technique is more simple than that of the Aldes' original technique, but its electron microscopic effect is same with that of the original technique.
