Smad 4基因在鼻咽癌CNE2细胞中的突变分析

摘要(Abstract):

目的:研究CNE2细胞中Smad4基因的突变。方法:运用RT-PCR方法从CNE2细胞中克隆人Smad4cDNA序列,然后与正常鼻咽组织及GenBank序列进行比较分析。结果:CNE2细胞中Smad4基因没有发生任何形式的突变。结论:CNE2细胞可能是一种非Smad4突变的恶性肿瘤,构建的含有人Smad4编码序列的重组质粒,为今后开展Smad4基因的功能研究奠定了基础。
Objective:To investigate the mutation of Smad 4 gene in CNE2 cell line.Methods:Human Smad 4 gene cDNA sequence was obtained from CNE2 cell by using RT-PCR technique,and compared with Smad 4 gene sequences obtained from normal nasopharynx tissue and GenBank.Results:No Smad 4 gene mutation was found in CNE2 cells.Conclusions:CNE2 cell is a kind of malignant tumor without mutation of Smad 4 gene.So,it is suggested that Smad 4 gene may not be possibly a suspectable factor for human nasopharyngeal carcinomas.The recombinant plasmid containing Smad 4 gene developed in this study lays a base for the functional research of Smad 4.
