燃煤型氟中毒对雌鼠卵巢TGF-α的影响

摘要(Abstract):

目的:动态观察燃煤型氟中毒对雌鼠卵巢转化生长因子(TGF-α)的影响。方法:SD雌鼠90只,随机均分为对照组、中氟组和高氟组,于不同时间段(60 d、120 d、180 d)观察氟斑牙,用离子选择电极法测定雌鼠尿氟、骨氟含量;电镜下观察卵巢凋亡情况,用TUNEL技术检测卵巢凋亡细胞数,用免疫组化法检测卵巢TGF-α的表达。结果:随着染氟剂量增加和染氟时间的延长,雌鼠氟斑牙症状逐渐加重,尿氟、骨氟含量增加,染氟组卵巢凋亡细胞数明显增加;与对照组相比,相同染氟时间各染氟组卵巢颗粒细胞TGF-α蛋白表达量均降低(P<0.05),高氟组低于中氟组,差异有统计学意义(P<0.05);相同染氟剂量,随着染氟时间延长,各染氟组TGF-α含量逐渐减少,中氟组60 d高于120 d和180 d,差异有统计学意义(P<0.05)。结论:氟中毒可能导致卵巢组织细胞超微结构损伤,严重破坏颗粒细胞中线粒体,影响卵巢细胞的能量供应,促使卵巢颗粒细胞凋亡;氟中毒可导致TGF-α分泌减少使其参与阻碍卵泡凋亡和闭锁的作用减弱,促使卵巢颗粒细胞的凋亡增加。
Objective: To investigate the influence of coal burning fluorosis on female rat's ovary TGF-α. Methods: Ninety female SD rats were divided into three groups randomly: control group,middle fluorine group and high fluorine group. At different period of time( 60 d,120 d,180 d) observing dental fluorosis,testing fluorine content of urine and bone with ion selective electrode method;The changes of ovary and granular cell apoptosis state were observed by transmission electron microscope; the apoptosis cell numbers in ovarian tissue were tested by Tunel method; the expression of TGF-α protein in ovaries of rats were detected by immunohistochemistry. Results: With the increase of fluoride dose and exposure time,the symptoms of dental fluorosis became worse,urinary fluorine and fluorine contents in bone increased,and the apoptosis cells in ovarian tissue was clearly on the rise in fluoride exposure groups. With the same exposure time,TGF-α protein contents in ovary granuler cells of fluoride exposure groups decreased comparing with control group( P < 0. 05),and those in high fluorine group were lower than those in middle fluorine group( P < 0. 05). With same amount of fluoride,increased exposure time reduced TGF-α levels gradually: middle fluorine group of 60 d were higher than those of 120 d and 180 d( P < 0. 05). Conclusions: Fluorosis may cause ovarian tissue structural damage and granular cell apoptosis and severely damage the mitochondrion,affecting the energy supply of ovarian cells. Fluorosis will lead to hypoeccrisis of TGF-α and weaken its effect of blocking apoptosis and interlock of ovarian follicle and promote the apoptosis of granule cells.
