某食用白酒对大鼠脑垂体ACTH阳性细胞表达及超微结构的影响Effects of Liquor on Expression and Ultrastructural Changes of ACTH Positive Cells of Pituitary in Rats

摘要(Abstract):
目的:研究某品牌食用白酒对大鼠脑垂体ACTH阳性细胞的表达及其超微结构的影响。方法:正常成年雄性SD大鼠,随机分为正常对照组及实验组,其中实验组分为4周组(4 w)、8周组(8 w)及12周组(12w)3个时间点,每个时间点又分为低剂量组[L组,0.8 m L/(kg·day)]、中剂量组[M组,1.6 m L/(kg·day)]及高剂量组[H组,2.4 m L/(kg·day)];各时间段实验大鼠均有5只作正常对照,实验组大鼠予相应食用白酒灌胃,每日11∶00 am及5∶00 pm各灌胃1次;正常对照组不予处理,分别于第4 w末、第8 w末,第12 w末取脑垂体组织;采用免疫组化SABC法、透射电镜技术、图像分析及形态计量法观察大鼠的脑垂体组织学改变、ACTH阳性细胞表达及平均光密度值。结果:与正常组比较,除4 w时H组ACTH阳性细胞表达有所升高之外,其余各组各时间点与正常对照组比较差异无统计学意义(P>0.05);透射电镜观察结果也表明,除4 w时H组大鼠ACTH阳性细胞超微结构稍有改变外,其余各组结构正常。结论:在本实验设定的剂量和时程内用某食用白酒灌胃,对大鼠垂体ACTH阳性细胞的表达和超微结构无明显影响。
Objective: To investigate the effect of liquor( 54% vol) on expression and ultrastructure ACTH positive cells of pituitary in rats. Method: Normal male adult SD rats were randomly divided into normal control group and experimental group. The experiment group were divided into 4 weeks group( 4 w),8 weeks group( 8 w) and 12 weeks group( 12 w). Each group of rats in different week group were further divided into low dose group [L dose,0. 8 m L /( kg·d) ],middle dose group [M dose,1. 6 m L /( kg·d) ]and high dose group 9H dose,2. 4 m L /( kg·d) ]. Rats in experimental group received intragastric administration of liquor twice a day( 11 am and 5 pm),while rats in normal control group received no special treatment. Pituitary glands were excised at the end of 4 weeks,8weeks and 12 weeks. Tissues were assessed by immunohistochemical SABC,transmission electron microscopy( TEM),image analysis and morphometry methods in the study. Result: Compared with the normal control group,there was no significant difference in the mean density of ACTH positive cells among groups( P > 0. 05) except the 4 weeks H group. The results of electron microscopy showed that the structure of ACTH positive cells in group were not changed except the 4 weeks H group. Conclusion: In the dose and time course of this experiment,the administration by gavage of this brand 54°consumption of liquor has no obvious effect on the expression and the ultrastructure of ACTH positive cells of pituitary in rats.
