免疫粘附玫瑰花试验条件初探
A PRIMARY SURVEY OF IMMUNE ADHERENCE ROSETTE TEST(IART)

摘要(Abstract):

本文利用正交设计试验法对影响免疫粘附玫瑰花(IAR)形成的诸多因素作了初步探讨。方差分析结果表明:抗体的量.补体浓度、致敏的温度和时间以及 O 型红细胞的来源等因素均对 IAR 的成花百分率有明显影响。文中对获得最佳结果的实验条件进行了讨论。该法能初步应用于检测杂交瘤细胞培养上清液中的单克隆抗体,特别适于检测带有 IgGFc 受体的抗原靶细胞上的表面抗原。
The factors which influences on IART were analysed primarily by ortho- gonal latin square design.The results of analysis suggested that some factors have obvious influence on the percentage of IAR such as the number of auti- bodies,the concentration of the complements,the Sensitizing duration and temperature,the origin of the human erythrocytes of group 0 and so on. The best result can be obtained by the following experimental condition: sensitizing the target cells with specific antibody,1/10 diluted guinea com- plement and the human erythrocytes of group 0 collected from different goners. This method has been used to examine and determine the McAb in the supernate of hybridoma cell cultures,the human peripheral lymphocytes and fetal thymocytes.All results are satisfactory.We think it is suitable to exa- mine surface antigens of cells bearing IgG Fc receptors.
