兴义维蚋气管鳃形态发育过程的初步研究Preliminary Study of the Morphological Development Process of Simulium xingyiense Tracheal Gill

摘要(Abstract):
目的:探究兴义维蚋幼虫期气管鳃形态发育的基本过程。方法:参考文献设计、建造蚋类实验室饲养系统,野外采集蚋卵、幼虫和蛹,用常规形态学和DNA条码技术鉴定蚋种,并在自行设计的饲养系统中饲养,每24 h收集幼虫;在显微成像系统下观察蚋幼虫各龄期气管鳃形态。结果:改进和建立了原代野生蚋类人工饲养系统,鉴定采集的蚋种为兴义维蚋,其1~5龄幼虫期无明显气管鳃分子化形态,6龄幼虫中可见囊状未分支气管鳃,7龄幼虫时气管鳃发生分支化,形成8条呼吸丝,分4对,其中5对呼吸丝透过幼虫表皮可见。结论:成功设计并建立了野生蚋类原代人工饲养系统,兴义维蚋1~5龄幼虫气管鳃呈透明团块状,6龄分支化形态为囊状未分支形态,7龄幼虫出现完全的分支化形态。
Objective: To study the basic morphological development process of black fly Simulium xingyiense tracheal gills. Methods: A Simuliid rearing system( SRS) were designed and established for collecting black fly larvae of arbitrarily stages. Simuliid eggs,larvae and pupae were collected in the field,identified based on morphology and DNA barcode sequence,and reared in the SRS. Larvae were collected every 24 hours. The morphology of tracheal gills of S. xingyiense was recorded under a microscopic imaging system. Results: An SRS was designed and constructed successfully. There existed no obvious tracheal gill during the larval instars 1 to 5 of S. xingyiense. In larval instar 6,the tracheal gill presents as a sac formation. In larval instar 7,the tracheal gill sac divided into 8 respiratory filaments,which forming 4 pairs,with 5 filaments can be observed through the larval cuticle. Conclusion: An SRS was designed and constructed successfully. The tracheal gill assumes transparent conglomeration during the larval instars 1 to 5 of S. xingyiense. In larval instar 6,the tracheal gill presents as a sac formation. In larval instar 7,the tracheal gill presents completely branching morphology.
